日本鳗鲡mavs基因的克隆及表达模式分析

摘要 利用PCR技术克隆获得了日本鳗鲡(Anguilla japonica)线粒体抗病毒信号蛋白(mitochondrial antiviral signaling, mavs)基因(Ajmavs),其开放阅读框有1368 bp,编码456个氨基酸。序列结构分析结果显示,Ajmavs含有3个保守结构域,分别为氨基端半胱天冬酶募集结构域、富含脯氨酸的结构域,以及羧基端跨膜结构域。系统发育树结果显示,Ajmavs与鱼类的mavs聚为一支,哺乳类、两栖类、鸟类及爬行类聚为一支。实时定量PCR(qPCR)结果显示,Ajmavs在日本鳗鲡各组织中均有表达,其中脑组织中的表达量最高,心脏其次,而性腺最低。用Poly I∶C(病毒RNA类似物)刺激12 h后,头肾、脾脏、肠组织中Ajmavs的表达水平显著增加,是对照组的21.31、30.33和15.96倍,暗示Ajmavs参与了日本鳗鲡的抗病毒免疫反应过程。 In this study,the mitochondrial antiviral signaling protein(mavs)encoding gene was cloned from the Japanese eel,Anguilla japonica(Ajmavs),using polymerase chain reaction.The transcript of Ajmavs contains an open reading frame(ORF)of 1368 bp,encoding 456 amino acids.Sequence structure analysis revealed the presence of three conserved structural domains within Aj mavs:the N-terminal caspase activation and recruitment domains(CARD),the proline-rich region(PRR),and the C-terminal transmembrane domain(TM).Phylogenetic analysis results showed that Aj mavs clustered with fish mavs,while mavs from mammals,amphibians,birds and reptiles clustered together.Gene expression results revealed that Ajmavs was expressed in all tissues examined,showing the highest levels in brain tissue,followed by the heart,and the lowest levels in the gonads.After 12 hours following stimulation of the viral RNA analogue Poly I:C,there was a significant increase in Ajmavs expression in the head kidney,spleen,and intestine,being 21.31,30.33 and 15.96 folds compared to the control group.Taken together,this results suggest an important role of Ajmavs in eel’s antiviral immune response.
