大黄素调节AGEs-RAGE信号通路对妊娠糖尿病大鼠的改善作用

摘要 目的:探讨大黄素对妊娠期糖尿病(GDM)大鼠糖脂代谢和炎症反应的改善作用及机制。方法:采用高糖高脂饮食合并注射链脲佐菌素方式建立GDM大鼠模型,将造模大鼠随机分为模型组(GDM组),大黄素低、中、高剂量组,二甲双胍组,另设置正常组。H-E染色和Annexin V-FITC/DAPI染色分别检测胰腺组织病理学变化和凋亡细胞;分别检测血液空腹血糖(FBG)、空腹胰岛素(FINS)、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)、肿瘤坏死因子-α(TNF-α)、白细胞介素(IL)-1β、IL-6、丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、铁离子水平及晚期糖基化终产物(AGEs)-晚期糖基化终产物受体(RAGE)通路蛋白表达。结果:与GDM组比较,二甲双胍组及大黄素各剂量组病理损伤好转,FBG、胰腺组织细胞凋亡率、血清TG、TC、LDL-C、TNF-α、IL-6、IL-1β水平、胰腺组织中MDA、铁离子水平及AGEs-RAGE通路蛋白表达水平下降,血清FINS,胰腺组织中SOD、GSH-Px水平上升。结论:大黄素可改善GDM大鼠的糖脂代谢和全身炎症反应,改善胰腺组织细胞凋亡,可能通过调节AGEs-RAGE信号通路实现。 Objective:To investigate the effects and mechanism of emodin in improving glucose and lipid metabolism and inflammatory response in rats with gestational diabetes mellitus(GDM).Methods:The model of gestational diabetes rats was established by combining a high sugar and high fat diet with streptozotocin injection.The model rats were randomly divided into model group(GDM group),low,medium and high dose emodin groups,and metformin group,with an additional normal control group.Histopathological changes and apoptosis of pancreatic cells were detected by H-E and Annexin V-FITC/DAPI staining.Fasting blood glucose(FBG),fasting insulin(FINS),the level of triglycerides(TG),total cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),highdensity lipoprotein cholesterol(HDL-C),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β),interleukin-6(IL-6),malondialdehyde(MDA),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px),iron ions,and the expression of advanced glycation end products(AGEs)-receptor for advanced glycation end products(RAGE)pathway proteins were detected,respectively.Results:Compared with the GDM group,the pathological injury of the metformin group and the emodin group was improved.The levels of FBG,the apoptosis rate of pancreatic tissue,serum TG,TC,LDL-C,TNF-α,IL-6,IL-1β,MDA,iron ion levels and AGES-AGE signal pathway protein expression were decreased significantly,while serum FINS,the level of SOD and GSH-Px in pancreatic tissue were increased significantly.Conclusion:Emodin can improve glucose-lipid metabolism and systemic inflammatory response in GDM rats,reduce pancreatic tissue apoptosis,and may achieve these effects by modulating the AGEs-RAGE signaling pathway.
