摘要
目的优化芹菜籽挥发油提取工艺,并研究其体外抗氧化活性。方法以芹菜籽挥发油提取率为考察指标,以药材粒度、微波功率、提取时间、液料比为考察因素,采用单因素试验结合响应面法,优化芹菜籽挥发油的微波辅助水蒸气蒸馏法提取工艺,并通过DPPH、ABTS及羟基自由基清除率试验研究其体外抗氧化活性。结果药材粒度为65目、微波功率为730 W、提取时间为46 min、液料比为23倍的工艺条件下,芹菜籽挥发油的提取率为(1.193±0.154)%,与响应面法预测值(1.190%)接近;体外抗氧化试验结果表明,芹菜籽挥发油具有一定的抗氧化活性,能够清除DPPH、ABTS和羟基自由基,其IC50分别11.722 mg/mL、10.426 mg/mL、8.645 mg/mL。结论微波辅助水蒸气蒸馏法操作简便易行、提取率高,为芹菜籽挥发油的开发利用提供理论依据。
Objective To optimize the extraction process of essential oil from celery seed and study its oxidative activity in vitro.Methods With the yield of essential oil of celery seed as the index,the particle size,microwave power,extraction time,and liquid-solid ratio were investigated as the influencing factors.A singlefactor experiment combined with response surface methodology was used to optimize the microwave-assisted steam distillation extraction process of apigenin essential oil from celery seeds.The anti-oxidant activity of the extract against DPPH free radicals,ABTS free radicals,and hydroxyl free radicals were studied.Results The results showed that the optimal extraction process for celery seed essential oil was a particle size of 65 mesh,microwave power of 730 W,extraction time of 46 minutes,and a liquid solid ratio of 23 times,and the extraction rate of celery seed essential oil was(1.193±0.154)%,which was close to the predicted value of response surface methodology(1.190%).The in vitro anti-oxidant activity test showed that the celery seed essential oil has a certain ability to scavenge DPPH free radicals,ABTS free radicals,and hydroxyl free radicals,and ICso values were 11.722 mg/mL,10.426 mg/mL and 8.645 mg/mL,respectively.Conclusion The microwave-assisted steam distillation method is simple and easy to operate,with high extraction efficiency,providing theoretical basis for the development and utilization of celery seed essential oil.
