摘要
多重PCR技术可在同一反应体系完成扩增反应实现对多个靶标的同步检测。随着多重PCR技术的发展,研究者们基于多重PCR扩增阶段优化引物设计,已开发出适用于第一、二、三代PCR技术的多重PCR方法。基于扩增产物检测阶段,并与琼脂糖凝胶电泳、毛细管电泳、色谱、质谱、荧光检测、纳米孔测序及生物传感技术相结合,研发出适用于多重PCR扩增产物的检测方法。多重PCR技术具有高通量、高效率的优势,得到了研究者的广泛关注,现该技术已被广泛应用于医学、畜牧、兽医、食品、农业等多个领域。本文综述了多重PCR技术的基本原理、分类和在动物疫病检测中的应用及发展前景,以期为多重PCR技术在兽医实验室动物疫病检测的推广提供参考。
Multiplex PCR technology can complete the amplification reaction in the same reaction system,combined with certain detection method to realized the detection of multiple targets.With the development of multiplex PCR technology,researchers have developed multiplex PCR methods applicable to the first,second and third generation of PCR technology based on the optimization of primer design in the amplification phase of multiplex PCR.Based on the detection of amplification products,they have developed methods applicable to the detection of multiplex PCR amplification products,combined with agarose gel electrophoresis,capillary electrophoresis,chromatography,mass spectrometry,fluorescence detection,nanopore sequencing and biosensing techniques and so on.Multiplex PCR technology has the advantages of high throughput and high efficiency,which has received extensive attention from researchers.It has been widely used in medicine,animal husbandry,veterinary,food,agriculture and other fields.In this paper,we reviewed the basic principle,classification,application and development prospect of multiplex PCR technology in animal epidemic disease detection,in order to provide reference for the promotion of multiplex PCR technology in animal disease detection in veterinary laboratories.
