谷瘟拮抗木霉筛选与鉴定及其防效研究

摘要 谷瘟病是谷子生产中的一种主要病害,严重影响谷子产量和品质。利用生防微生物防治谷瘟病是未来绿色植保的发展趋势。以河北省农林科学院谷子研究所石家庄郄马试验站谷子根际土壤为试验材料,利用孟加拉红培养基分离木霉菌株,采用平板对峙法筛选最优菌株,通过形态学观察与Tef1序列分析相结合的方法进行菌种鉴定,利用盆栽试验明确菌株对谷瘟病的防治效果。结果表明:从谷子根际土壤样品中共筛选到菌落形态具有木霉典型特征的菌株24株,其中KE-1菌株的拮抗效果最好,对谷瘟病菌、玉米大斑病菌、玉米小斑病菌、禾谷镰孢、拟轮枝镰孢、灰霉的抑菌率分别为42.86%、44.44%、57.89%、53.33%、60.00%和42.22%。综合形态特征及Tef1序列分析结果,最终确定KE-1菌株为哈茨木霉(Trichoderma harzianum)。盆栽试验结果显示,用哈茨木霉孢悬液进行谷子灌根后,谷子发病程度明显减轻,对谷瘟病菌防效达到55.56%。哈茨木霉菌株KE-1具有较好的生防潜力,在谷瘟病害的生物防治中具有潜在的应用价值。 Pyricularia grisea is a major disease in millet production,severely affecting the yield and quality of millet.The use of biocontrol microorganisms to prevent and control P.grisea is the development trend of green plant protection in the future.Using the rhizosphere soil of millet at Shijiazhuang Qiema Experimental Station of the Millet Research Institute of Hebei Academy of Agriculture and Forestry Sciences as the experimental material,Trichoderma strains were isolated using Rose Bengal medium.The optimal strains were screened using the plate confrontation method,and the strains were identified through a combination of morphological observation and Tef1 sequence analysis.The efficacy of the strains against P.grisea was determined through pot experiments.The results showed that a total of 24 strains with typical characteristics of Trichoderma were screened from the rhizosphere soil samples of millet.Among them,KE-1 strain had the best antagonistic effect,with the inhibition rates of 42.86%,44.44%,57.89%,53.33%,60.00%,and 42.22%against P.grisea,Setosphaeria turcica,Bipolaris maydis,Fusarium graminearum,F.verticil-lioides and Botrytis cinerea,respectively.Based on the comprehensive morphological characteristics and Tef1 sequence analysis results,KE-1 strain was ultimately determined to be Trichoderma harzianum.The results of the pot experiment showed that after root irrigation with T.harzianum suspension,the disease severity of millet was significantly reduced,with a control efficacy of 55.56%against the pathogen of P.grisea.In conclusion,T.harzianum strain KE-1 has good biocontrol potential and holds potential application value in the biological control of P.grisea.
