甜茶外植体消毒效果及初代诱导培养

摘要 为解决甜茶[Rubus chingii var.suavissimus(S.Lee)L.T.Lu]组培苗消毒过程中容易出现污染及初代诱导困难的问题,分别以甜茶的顶芽、带芽茎段、半木质化茎段为外植体,研究甜茶外植体的最佳消毒方法、最佳外植体及初代诱导的适宜条件。结果表明,甜茶最佳外植体为顶芽,最佳消毒处理为75%乙醇浸泡30 s,0.1%升汞消毒10 min;顶芽污染率最低,仅为20.83%,出芽率最高,为70.83%;最佳初代诱导生长调节剂组合为MS+TDZ 1.0 mg/L+NAA 0.2 mg/L,芽诱导率最高,为66.67%。 In order to solve the problems of contamination and difficulty in initial induction during the disinfection process of sweet tea[Rubus chingii var.suavissimus(S.Lee)L.T.Lu]tissue culture seedlings,the optimal disinfection method,optimal explants,and suitable conditions for initial induction of sweet tea explants were studied using the apical bud,bud bearing stem segment,and semi lignified stem segment as explants.The results showed that the optimal explant for sweet tea was the apical bud,and the optimal disinfection treatment was soaking in 75%ethanol for 30 seconds and disinfecting with 0.1%mercuric chloride for 10 minutes;the top bud pollution rate was the lowest,only 20.83%,and the germination rate was the highest,at 70.83%;The optimal combination of primary induction growth regulators was MS+TDZ 1.0 mg/L+NAA 0.2 mg/L,with the highest bud induction rate of 66.67%.
