手术关胸时间对大鼠急性心肌梗死模型存活率的影响

The Effect of Different Thoracic Closure Time Points on the Survival Rate of Acute Myocardial Infarction in Rat Model

摘要 左冠状动脉前降支(LAD)结扎后不同时间点进行心肌注射使得手术关胸时间产生差异,探究手术关胸时间差异对急性心肌梗死(AMI)大鼠存活率的影响,旨在为心肌梗死模型药物干预试验提供参考依据。选取24只雄性SD大鼠,随机分为4组,每组6只。其中对照组进行假手术,即只过针不结扎;3个处理组,分别在LAD结扎后的10、30、45 min于左心室心肌定点注射生理盐水。连续记录21 d内处理组和对照组的大鼠存活天数并计算大鼠存活率。结果显示,LAD结扎后,3个处理组心电图ST段明显抬高,21 d后左心室前壁变薄;21 d内10 min注射组的大鼠存活率高达50%。结果表明,LAD结扎后应尽快注射,即手术关胸时间越早,术后大鼠存活率越高。研究结果可为LAD结扎后的注射时间提供参考范围,并为AMI相关干预研究提供依据。 Myocardial injection at different time points after LAD ligation makes the time of closing the chest different.To explore the effect of the time difference of closing the chest on the survival rate of AMI rats,and to provide reference for the drug intervention experiment of myocardial infarction model,24 male SD rats were randomly divided into 4 groups with 6 rats in each group.The control group was subjected to sham operation,that is,only the needle was passed without ligation;the three treatment groups were injected with normal saline at the left ventricular myocardia at 10 min,30 min,and 45 min after LAD ligation,respectively.The survival days of the rats were calculated continuously.The results showed that,after ligation of LAD,the ST segment of ECG was significantly elevated in the three treatment groups,and the anterior wall of the left ventricle became thinner after 21 days.Within 21 days,the survival rate of rats in the 10 min injection group was up to 50%.Therefore,the LAD should be injected as soon as possible after ligation,that is,the earlier the time of thoracic closure,the survival rate of rats after operation showed an upward trend.This study can provide a reference range for the injection time after LAD ligation and provide a basis for AMI related intervention research.
