摘要
目的探讨大鼠急性心肌梗死后不同时间点心肌组织中p38mRAN、p38蛋白的表达水平及意义。方法健康成年雄性SD大鼠36只,将其随机分为对照组、假手术组和心肌梗死模型组,模型组大鼠开胸结扎冠状动脉前降支以建立心肌梗死模型,成功结扎冠状动脉前降支后1、3、6、12h无痛处死大鼠并获取大鼠心肌组织;对照组大鼠不做任何处理;假手术组大鼠开胸冠状动脉穿线但不结扎冠状动脉;术后均无痛处死并获取心肌组织;分别采用RT-PCR、Western blot测定大鼠心肌组织p38mRNA及蛋白的表达水平。结果对照组与假手术组之间p38mRNA及蛋白表达的差异均无统计学意义(P>0.05);AMI组大鼠各亚组心肌组织中p38mRNA及蛋白表达均高于假手术组(P<0.05);AMI组中不同时间点亚组大鼠心肌组织中p38mRNA及蛋白的表达差异均有统计学意义(P<0.05),其表达量随结扎冠状动脉前降支时间的延长而增加,并于6h表达量达到高峰,随后下降,至12h其表达水平与假手术组差异无统计学意义(P>0.05)。结论 p38可能在心肌梗死后的病理过程中发挥作用。
Objective To investigate the expression of P38mRNA and P38 mitogen-activated protein kinase(MAPK) after acute myocardial infarction(AMI) in different time and to explore its changing trend.Methods 36 healthy adult male Sprague-Dawley rats were equally and randomly divided into model groups(including 4 subgroups),sham operation groups and blank control groups.In the model groups,Rats' anterior descending coronary were ligated to established the model of acute myocardial infarction(AMI).Rats were sacrificed after ligating 1 hour,3 hour,6 hour and 12 hour respectively in each subgroups and collecting the heart tissue.The rats in sham operation group were opened chest but didn't ligate the anterior descending coronary;The black group rats were given the normal diet and no special treating.After the end of experiment the sham operation group and blank control group rats were sacrificed and obtained their heart tissue.Results The expression of P38 mRNA and P38 between the sham operation group and blank control group showed no significantly different(P > 0.05).But between the AMI group and the sham operation group,there was significantly different in expression(P < 0.05).The expression of p38mRNA and p38 in each subgroup of AMI group were significantly different(P < 0.05).After ligating anterior descending coronary 1 hour,3 hour,6 hour and 12 hour,the expression raised gradually and the maximum expression was at 6 hour,then gradually descending to the normal level and at this time,the difference between AMI group and sham operation group was no significant(P > 0.05).Conclusion Expression of P38 in the heart tissue of rats may participate in pathological process after AMI.
