摘要
目的研究低温等离子照射治疗SD大鼠皮肤创伤。方法SPF级健康雄性SD大鼠42只简单随机法分组为5组,对照组(WD)、低温等离子组(D)、赛霉安散组(ZZ)、联合治疗组(DZ)每组9只,正常组(CD)6只。WD组、D组、ZZ组、DZ组大鼠麻醉后刮去背部毛发在背部正中延脊柱划开1 cm长伤口,CD组大鼠仅刮去背毛。每天记录大鼠体重、伤口状态,用乳酸依沙吖啶溶液处理创面,D组对伤口进行1 min低温等离子(Low-temperature Plasma,LTP)照射,ZZ组用赛霉安散涂抹治疗,DZ组涂抹赛霉安散治疗并辅以1 min LTP照射,WD组及CD组不做其他处理。分别取治疗第3天、7天、14天创面处皮肤组织,制作石蜡组织切片进行HE染色,观察伤口不同恢复期形态变化。取治疗第7天伤口皮下肌肉组织,组织匀浆后使用试剂盒测试白介素1(IL-1)及谷胱甘肽含量。结果D组大鼠恢复期生长速度略慢但与其他组大鼠体重无显著性差异。HE染色结果显示,治疗3 d时各组恢复无显著差异,7 d时D组、ZZ组、DZ组明显好转并且DZ组效果更好,14 d时各组基本恢复,DZ组瘢痕更少。7 d时伤口皮下肌肉组织WD组大鼠IL-1表达与CD组相比,有显著性差异(P<0.001),与WD组相比较各治疗组IL-1表达均有显著性改善(P<0.001),DZ组趋势更明显;WD组大鼠GSH含量与CD组相比,有显著性降低(P<0.05),D组和DZ组与WD组相比GSH含量显著增加(P<0.05),WD组大鼠GSSG含量与CD组相比,有显著性增加(P<0.05),DZ组与WD组相比GSSG含量显著降低(P<0.05)。结论LTP照射可抑制炎症因子渗出,加速皮肤抗炎、抗氧化剂和自由基清除,促进大鼠皮肤创伤愈合,减少瘢痕的形成。
Objective This study aimed to investigate the therapeutic effects of low-temperature plasma(LTP)irradiation on skin wounds in SD rats.Methods A total of 42 SPF-grade healthy male Sprague-Dawley(SD)rats were randomly assigned to five groups:the control group(WD),the low-temperature plasma group(D),the Saimei’an Powder group(ZZ),and the combination therapy group(DZ),with 9 rats in each group,while the normal group(CD)consisted of 6 rats.Rats in the WD,D,ZZ,and DZ groups were anesthetized,and after shaving the dorsal hair,a 1 cm-long incision was made along the midline of the back.In the CD group,only dorsal hair shaving was performed without incision.Body weight and wound condition were recorded daily.The wounds were treated with ethacridine lactate solution for infection prevention.In the D group,the wound was exposed to LTP irradiation for 1 minute;in the ZZ group,Saimei’an Powder was applied to the wound;in the DZ group,Saimei’an Powder was applied in combination with 1-minute LTP irradiation.The WD and CD groups received no additional treatment.On days 3,7,and 14 post-treatment,skin tissue from the wound site was collected,embedded in paraffin,and subjected to hematoxylin and eosin(HE)staining to assess morphological changes at different stages of wound healing.On day 7 post-treatment,subcutaneous muscle tissue from the wound area was harvested,homogenized,and analyzed using assay kits to determine the levels of interleukin-1(IL-1)and glutathione(GSH).Results During the recovery period,the growth rate of rats in the D group was slightly slower;however,no statistically significant difference in body weight was observed compared to the other groups.HE staining showed no notable differences in wound healing among the groups on day 3 post-treatment.By day 7,the wounds in the D,ZZ,and DZ groups exhibited significant improvement,with the DZ group showing the most pronounced effect.By day 14,all groups had largely recovered,with the DZ group displaying less scarring.On day 7,interleukin-1(IL-1)expression in the subcutaneous muscle tissue of the wound site was significantly higher in the WD group compared to the CD group(P<0.001).However,IL-1 expression was significantly reduced in all treatment groups compared to the WD group(P<0.001),with the most pronounced reduction in the DZ group.Additionally,GSH levels in the WD group were significantly lower than those in the CD group(P<0.05).In contrast,GSH levels in the D and DZ groups were significantly higher than those in the WD group(P<0.05).Moreover,oxidized glutathione(GSSG)levels in the WD group were significantly elevated compared to the CD group(P<0.05),whereas the DZ group exhibited a significant reduction in GSSG levels compared to the WD group(P<0.05).Conclusion LTP irradiation can inhibit inflammatory cytokine infiltration,enhance anti-inflammatory and antioxidant responses,and promote wound healing while reducing scar formation in SD rats.
