鱼腥草挥发油对佐剂性膝关节骨性关节炎大鼠血清MMP-3、COX-2、SOD表达的影响

摘要 目的:探讨鱼腥草挥发油对佐剂性膝关节骨性关节炎(KOA)大鼠血清基质金属蛋白酶-3(MMP-3)、环氧合酶-2(COX-2)、超氧化物歧化酶(SOD)表达的影响。方法:将SD健康大鼠随机分为空白对照组(正常组)、KOA模型对照组(模型组)、阳性药物对照组(药物组)以及鱼腥草挥发油低、中、高剂量组,每组10只,雌雄各半;注射弗氏完全佐剂进行大鼠模型制备,建模成功后,药物组灌胃盐酸氨基葡萄糖水溶液,低、中、高剂量组灌胃鱼腥草挥发油制备液,正常组和模型组灌胃生理盐水,1次/d,持续6周。ELISA法测定各组血清MMP-3、COX-2、SOD表达及TNF-α、IL-8、IL-10水平,观察膝关节滑膜组织病理改变。结果:与正常组比较,模型组和各灌胃组大鼠血清TNF-α、IL-8水平及MMP-3、COX-2表达显著升高(P<0.05),IL-10水平、SOD表达显著下降(P<0.05),与模型组比较,各灌胃组大鼠血清TNF-α、IL-8水平及MMP-3、COX-2表达显著降低(P<0.05),IL-10水平、SOD表达显著升高(P<0.05),其中高剂量组各项观察指标与药物组比较,差异无统计学意义(P>0.05),病理学观察显示,高剂量组的炎症缓解程度更明显,接近于药物组。结论:鱼腥草挥发油能够缓解佐剂性KOA大鼠炎症,其作用机制可能与抑制MMP-3、COX-2表达,上调SOD表达有关。 Objective:To investigate the effect of volatile oil from Houttuynia cordata on the expression of serum matrix metalloproteinase-3(MMP-3),cyclooxygenase-2(COX-2),and superoxide dismutase(SOD)in adjuvant induced knee osteoarthritis(KOA)rats.Methods:SD healthy rats were randomly divided into blank control group(normal group),KOA model control group(model group),positive drug control group(drug group),and low,medium,and high dose groups of Houttuynia cordata volatile oil,with 10 rats per group,half male and half female.The rat model was prepared by injecting Freund’s complete adjuvant.After successful modeling,the drug group was orally administered with hydrochloric acid glucosamine aqueous solution,the low,medium,and high-dose groups were orally administered with volatile oil of Houttuynia cordata preparation solution,and the normal group and model group were orally administered with physiological saline once a day for 6 weeks.ELISA was used to measure the levels of serum MMP-3,COX-2,SOD,TNF-α,IL-8,and IL-10 in each group,and pathological changes in knee joint synovial tissue were observed.Results:Compared with the normal group,the levels of TNF-α,IL-8,MMP-3,and COX-2 in the serum of rats in the model group and each gavage group were significantly increased(P<0.05),while the levels of IL-10 and SOD were significantly decreased(P<0.05).Compared with the model group,the levels of TNF-α,IL-8,MMP-3,and COX-2 in the serum of rats in each gavage group were significantly decreased(P<0.05),while the levels of IL-10 and SOD were significantly increased(P<0.05).Among them,the observation indicators in the high-dose group were not statistically significant compared with the drug group(P>0.05).Pathological observations showed that the inflammation relief in the high-dose group was more obvious,close to the drug group.Conclusion:The volatile oil of Houttuynia cordata can alleviate inflammation in adjuvant induced KOA rats,and its mechanism of action may be related to the inhibition of MMP-3 and COX-2 expression and upregulation of SOD expression.
