北柴胡茎叶化学成分及其生物活性研究 被引量:2

摘要 采用薄层色谱、硅胶柱色谱、制备型高效液相色谱等多种色谱分离技术,对北柴胡茎叶中的化学成分进行分离,根据理化性质及波谱数据对结构进行鉴定。采用Ellman法测定化合物的乙酰胆碱酯酶抑制活性,并利用MTT法检测其对神经细胞的保护活性。从北柴胡茎叶中分离得到15个化合物,Erigeroflavanone(1),槲皮素(2),槲皮素-3-鼠李糖苷(3),山柰酚3-O-α-L-鼠李糖甙(4),木犀草素(5),Morin-3-O-α-rhamnopyranoside(6),咖啡酸甲酯(7),焦谷氨酸甲酯(8),3,4-二羟基苯甲酸甲酯(9),香草酸(10),3,4-二羟基苯甲酸(11),对苯二酚(12),1,2,4-苯三酚(13),9(Z)-octadecenamide(14)(Z)-21-methyl-8-pentatriacontene(15)。化合物1,4,6,8,12,14,15为首次从柴胡属植物中分离得到。化合物5~9,11,13~15具有乙酰胆碱酯酶抑制活性,IC50值分别为78.92μM、9.60μM、20.15μM、30.99μM、0.76μM、0.17μM、60.25μM、12.70μM、9.53μM。化合物1,3,5,9~12显示出神经细胞保护作用,最高细胞存活率分别为58.45%、56.88%、60.98%、84.4%、62.50%、60.43%、59.58%,其中化合物9神经保护作用最强。 Various chromatographic techniques,such as thin-layer chromatography,silica gel column chromatography,and preparative high-performance liquid chromatography were used to separate compounds from the stems and leaves of Bupleurum chinense DC.The structures of the obtained compounds were identified based on physicochemical properties and spectral data.The Ellman method was used to determine acetylcholinesterase activity,and the MTT method was used to determine neuroprotective activity.15compounds were isolated from Bupleurum chinense DC,erigeroflavanone(1),quercetin(2),quercetin-3-rhamnoside(3),kaempferol-3-O-α-L-rhamnoside(4),luteolin(5),morin-3-O-α-rhamnopyranoside(6),caffeic acid methyl ester(7),L-pyroglutamic acid methyl ester(8),protocatechuic acid methyl ester(9),vanillic Acid(10),protocatechuic acid(11),hydroquinone(12),1,2,4-benzenetri(13),9(Z)-octadecenamide(14),(Z)-21-methyl-8-pentatriacontene(15).Compounds 1,4,6,8,12,14,15 were isolated for the first time from the genus Bupleurum.The compounds numbered 5~9,11,13~15 exhibited acetylcholinesterase inhibitory activity,with IC_(50)values of 78.92μM,9.60μM,20.15μM,30.99μM,0.76μM,0.17μM,60.25μM,12.70μM,and 9.53μM.Compounds 1,3,5,9~12 demonstrated neuroprotective effects,with maximum neuronal survival rates of 58.45%、56.88%、60.98%、84.4%、62.50%、60.43%、59.58%,Among these,compound 9 exhibited the strongest neuroprotective effect.
