90份花生种子萌发期抗逆性鉴定与遗传多样性分析

摘要 以90个花生品种(系)种子为材料,分别经盐(0.12 mol/L NaCl)、碱(0.10 mol/L NaHCO 3,pH=8.3)、干旱(15%PEG-6000)、低温(2℃浸种72 h)胁迫处理后进行常温发芽,测定发芽势、发芽率、发芽指数、活力指数等4项指标,运用基于主成分分析的隶属函数法对参试花生芽期抗逆性进行综合评价。结果表明,抗逆性排名前十的品种(系)为HY666、21S400、21L186、HY669、21L119、HY9625、21L123、21L192、HY6620和HY9623。选用37对AhTE引物对参试花生进行遗传多样性分析,90份材料遗传相似系数均值为0.7942,在遗传相似系数为0.7840时,可将所有材料分为四大组。A组由高油酸材料和油酸含量较高的材料组成,B组可进一步分为B1和B2两个亚组,B1组多为高油酸花生,B2组多是高产普通花生,不同亚组包含的材料数差异较大。在综合抗性评价中,高耐材料主要分布在B组类群中,敏感材料主要分布在A组类群,中耐和低耐群体遗传多样性较丰富。 The seeds of 90 Arachis hypogaea varieties(systems)were treated with salt(0.12 mol/L NaCl),alkali(0.1 mol/L NaHCO 3,pH=8.3),drought(15%PEG-6000)and low temperature(2℃for 72 hours),respectively.The germination potential,germination rate,germination index and vitality index were determined.The resistance of flowering buds was evaluated by means of the genus function method based on principal component analysis.The results showed that the top ten products(lines)of reverse resistance are HY666,21S400,21L186,HY669,21L119,HY9625,21L123,21L192,HY6620 and HY9623.37 pairs of AhTE primers were used to analyze the genetic diversity of Arachis hypogaea samples.The average genetic similarity coefficient of 90 materials was 0.7942,and all materials could be divided into four groups when the genetic similarity coefficient was 0.7840.Group A was composed of high-oleic acid materials and materials with high oleic acid content,and group B could be further divided into two subgroups B1 and B2.Group B1 was mostly high-oleic acid peanuts,and group B2 was mostly high-yielding common peanuts.The number of materials contained in different subgroups varied greatly.In the comprehensive resistance evaluation,the high-tolerance materials were mainly distributed in group B,the sensitive materials were mainly distributed in group A,and the genetic diversity of the medium-tolerance and low-tolerance populations was higher.
