摘要
文章以奶牛乳腺上皮细胞为试验模型,探究叶酸对奶牛乳腺上皮细胞增殖及脂滴、β-酪蛋白合成的影响。用不同浓度的叶酸纯化处理奶牛乳腺上皮细胞,分别采用CCK-8法和Edu法检测奶牛乳腺上皮细胞增殖,筛选出叶酸最佳使用浓度。采用尼罗红法和组织细胞甘油三酯含量酶法检测不同处理组胞内胞外甘油三酯含量,通过Western blotting检测不同处理组β-酪蛋白(CSN2)及相关蛋白表达情况。结果表明,2.50μg/mL叶酸是促进奶牛乳腺上皮细胞增殖的最优浓度;叶酸可显著增加奶牛乳腺上皮细胞胞内胞外甘油三酯含量,显著提高CSN2蛋白表达,p-Akt、Akt、p-mTOR、mTOR、SREBP1、CyclinD1等蛋白表达显著提高。
To explore the effects of folic acid on synthesis of lipid droplet andβ-casein,dairy cow mammary epithelial cells were used as experimental models.Firstly,the mammary epithelial cells of dairy cows were treated with folic acid with different concentrations,and the proliferation of mammary epithelial cells of dairy cows was detected by CCK-8 method and Edu method to choose the best concentration of folic acid.Then the content of intracellular and extracellular triglycerides in different treatment groups was detected by Nile red staining and enzymatic method of triglyceride content in tissue cells.The results showed that 2.50μg/mL folic acid was the best concentration to promote the proliferation of mammary epithelial cells in dairy cows.Folic acid can significantly increase the intracellular and extracellular triglyceride content of cow mammary epithelial cells,significantly increase the expression of CSN2 protein,and significantly increase the expression of p-Akt,Akt,p-mTOR,mTOR,SREBP1,CyclinD1 and other proteins.
