摘要
筛选桔梗-甘草药对干预脂多糖(LPS)诱导小鼠急性肺损伤的最佳配伍比例。将96只雄性昆明小鼠随机平均分为空白对照组、模型组、地塞米松阳性药物组以及5个桔梗-甘草配伍干预组,连续给药7 d,最后1次给药1 h后,除空白对照组外,其他组均用LPS滴鼻法建立小鼠急性肺损伤模型。通过血常规、肺W/D比、脏器指数、TNF-α、IL-1β和IL-6水平、肺组织形态学观察,筛选出最佳配伍比例。结果表明,与空白对照组相比,模型组W/D比、血常规指标、IL-1β、TNF-α、IL-6差异显著(P<0.01或P<0.05);与模型组相比,桔梗-甘草配伍干预组(1∶2)W/D比、白细胞、血小板总数、IL-6、TNF-α、IL-1β差异显著(P<0.01或P<0.05);HE染色染色结果显示,模型组肺组织大量炎性细胞浸润,空白对照组肺泡结果清晰。说明桔梗-甘草配伍干预组(1∶2)对小鼠急性肺损伤干预效果最好。
The study aims to screen the optimal combination ratio of Platycodon grandiflorus-Glycyrrhiza for intervention in LPS induced acute lung injury in mice.96 male KM mice were randomly divided into a blank control group,a model group,a dexamethasone positive drug group,and five different compatibility groups of Platycodon grandiflorus-Glycyrrhiza,with 12 mice in each group.The mice were continuously administered for 7 days.After the last dose for 1 hour,except for the blank control group,all other groups were used to establish acute pneumonia models with LPS nose drop method in mice.The optimal compatibility ratio was screened through blood routine,lung W/D ratio,organ index,and TNF-α,IL-1β,IL-6 content detections and lung tissue morphology observation.The results showed that,compared with the blank control group,the W/D ratio,blood routine indicators,and IL-1β,TNF-α,IL-6 were significantly different in the model group(P<0.01 or P<0.05);Compared with the model group,the intervention group with the combination of Platycodon grandiflorus-Glycyrrhiza(1∶2)had significant differences in W/D ratio,white blood cells,total platelet count,IL-6 and TNF-α,IL-1β(P<0.01 or P<0.05);HE staining observed obvious inflammation in lungs of the model group,indicating the combination of Platycodon grandiflorus and Glycyrrhiza(1∶2)has the best intervention effect on acute lung injury in mice.
