基于网络药理学和分子对接技术的马齿苋治疗骨关节炎的作用机制研究 被引量:1

摘要 利用网络药理学探讨马齿苋(Portulaca oleracea L.)治疗骨关节炎(osteoarthritis,OA)的相关作用机制.首先,利用中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)数据库获取马齿苋的活性成分及作用靶点,并通过Uniprot数据库对靶点基因进行校正;其次,从GeneCards、DisGeNET数据库筛选骨关节炎的疾病靶点,并结合STRING平台、GO功能与KEGG通路富集分析马齿苋治疗骨关节炎的作用机制;最后,分子对接实验探究马齿苋活性成分与骨关节炎靶蛋白的亲和力与结合模式.结果初步表明马齿苋能够通过多成分、多靶点及多通路机制对骨关节炎进行干预.这一发现为后续深入探讨马齿苋在骨关节炎治疗中的应用奠定了理论基础. To explore the related mechanism of Portulaca oleracea L.in the treatment of osteoarthritis(OA)by network pharmacology.Firstly,the active components and targets of Portulaca oleracea L.were obtained by using the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)database,and the target genes were corrected by Uniprot database.Secondly,the disease targets of osteoarthritis were screened from GeneCards and DisGeNET databases,and the mechanism of Portulaca oleracea L.in the treatment of osteoarthritis was analyzed by combining STRING platform,GO function and KEGG pathway enrichment,Finally,molecular docking experiments were conducted to explore the affinity and binding mode of the active components of Portulaca oleracea L.with osteoarthritis target proteins.In summary,based on the systematic retrieval literature,the results preliminarily show that Portulaca oleracea L.can intervene in osteoarthritis through multi-component,multi-target and multi-pathway mechanisms.This finding laid a theoretical foundation for the further study of the application of Portulaca oleracea L.in the treatment of osteoarthritis.
