鸡蛋血肉斑性状全基因组关联分析

Genome-Wide Association Analysis of Blood-spotted Egg Traits in Chicken

摘要 为深入了解血肉斑性状的遗传基础,试验选用1200只28周龄洛岛红母鸡作为试验素材,测定了主要生产性状指标和包括血肉斑性状在内的蛋品质性状,从中选出连续3 d产血肉斑蛋和产无血肉斑蛋的鸡各60只,采血提取DNA,利用10×全基因组重测序,基于全基因组重测序和血肉斑表型数据进行全基因组关联分析。结果显示:产血肉斑蛋组和不产血肉斑蛋组的生产性状和蛋品质性状无显著差异(P>0.05);血肉斑性状遗传力为0.13;与血肉斑蛋性状相关的SNPs位点有303个,分布于1、2、3、4、5、7、9、10、11、12、13、14、16、19、25、26、31、33、40、41号染色体上,筛选到ATF4、CD47、FGF14、LSAMP、MAP4K4、PDGFB等重要候选基因。研究表明,血肉斑性状为中低遗传力,受微效多基因影响,其中ATF4、CD47、FGF14、LSAMP、MAP4K4、PDGFB为重要候选基因。 In order to understand the genetic basis of blood spot trait,1200 Rhode Island Red aged 28 weeks were selected as the experimental material,and the main production traits and egg quality traits including blood spot trait were measured.Blood samples were collected for DNA extraction,and 10×whole genome resequencing was used for genome-wide association analysis based on whole genome resequencing and blood stain phenotype data.The results showed that there was no significant difference in production traits and egg quality traits between the blood-spotted egg group and the bloodless-spotted egg group(P>0.05).The heritability of blood spot trait was 0.13.There were 303 SNPs associated with the blood-spotted egg trait,distributed on chromosome 1,2,3,4,5,7,9,10,11,12,13,14,16,19,25,26,31,33,40 and 41.ATF4,CD47,FGF14,LSAMP,MAP4K4,PDGFB and other important candidate genes were screened.The results indicated that blood-spotted traits had medium to low heritability and were affected by minor effect multiple genes.Among them,ATF4,CD47,FGF14,LSAMP,MAP4K4 and PDGFB were important candidate genes.
