基于PI3K/AKT/mTOR信号通路探讨理血解毒方对银屑病大鼠干预的机制研究 被引量:1

Exploring the intervention mechanism of Lixue Jiedu Decoction on psoriasic rats based on the PI3K/AKT/mTOR signaling pathway

摘要 目的基于PI3K/AKT/mTOR信号通路,通过对银屑病大鼠模型皮损指数进行评价,探索理血解毒方对银屑病的作用机制。方法将30只SD大鼠随机分为空白组、模型组、甲氨蝶呤西药对照组、理血解毒方中药低、中、高剂量组。局部使用5%的咪喹莫特乳膏构建银屑病大鼠模型,以各组相应药物对银屑病大鼠模型进行灌胃,计算皮损指数,通过ELISA、RT-PCR和Western Blot检测PI3K/AKT/mTOR信号通路血清因子、皮损基因表达以及皮损蛋白含量,分析各项指标与皮损指数间的相关性。结果与空白组比较,模型组大鼠出现典型的银屑病皮损。与模型组比较,甲氨蝶呤组、理血解毒方中药低、中、高剂量组中,大鼠PI3K/AKT/mTOR信号通路血清因子、皮损基因表达以及皮损蛋白含量均降低(P<0.05),其中,理血解毒方中药高剂量组与甲氨蝶呤西药对照组结果最为显著(P<0.05)。ΔMPASI与PI3K/AKT/mTOR信号通路各项指标呈负相关性(P<0.01)。结论理血解毒方可能通过抑制PI3K/AKT/mTOR信号通路的表达,发挥对银屑病的治疗作用。 Objective Based on the PI3K/AKT/mTOR signaling pathway,the skin lesion indexes of the psoriasis rat model were evaluated to explore the action mechanism of Lixue Jiedu Decoction(LJD)on psoriasis.Methods Thirty SD rats were randomly divided into the following groups:the blank group,the model group,the methotrexate control group,and LJD low-dose(LJD-L),medium-dose(LJD-M),and high-dose(LJD-H)groups.The psoriasic rat model was induced by topical application of 5%imiquimod cream,followed by treatment with the corresponding drugs in each group via gavage.The skin lesion indexes were calculated,and the serum factors of PI3K/AKT/mTOR signaling pathway,lesional gene expression,and lesional protein content were assessed using ELISA,RT-PCR,and Western blot(WB)techniques.The correlation between these factors and the skin lesion indexes were analyzed.Results Compared with the control group,the model group exhibited typical psoriatic skin lesions.Compared with the model group,the MTX group and the LJD-L,LJD-M and LJD-H groups showed significant reduction in serum factors of the PI3K/AKT/mTOR signaling pathway,gene expression of skin lesions,and skin lesion protein content(P<0.05).The LJD-H group and the methotrexate control group demonstrated the most remarkable results(P<0.05).Each index of Delta MPASI was negatively correlated with PI3K/AKT/mTOR signaling pathways(P<0.01).Conclusion LJD may play a therapeutic role in psoriasis via inhibiting the expression of PI3K/AKT/mTOR signaling pathway.
