摘要
目的探讨黄芩素与阿奇霉素联合喹诺酮类药物对多重耐药铜绿假单胞菌生物膜的抑制效果。方法随机选取10株多重耐药铜绿假单胞菌,通过微量肉汤稀释法测定黄芩素、左氧氟沙星、环丙沙星及阿奇霉素单独及联合用药的最小抑菌浓度(MIC)值,并计算部分抑菌指数(FICI)以评估联合用药效果。同时,采用结晶紫染色法评估药物对生物膜的抑制作用。结果黄芩素与左氧氟沙星、环丙沙星联合使用时表现出协同或相加作用。喹诺酮药物与黄芩素联合使用对10株铜绿假单胞菌的生物膜均显示出抑制效果;阿奇霉素与低浓度左氧氟沙星、环丙沙星联合使用时,对生物膜的抑制作用差异具有统计学意义(P<0.05)。结论中西药联合用药的效果优于单独用药。左氧氟沙星和环丙沙星能抑制铜绿假单胞菌生物膜的生长,且与黄芩素联合时对生物膜的抑制作用无显著差异。阿奇霉素能增强低浓度喹诺酮类药物的疗效。
Objective To investigate the inhibitory effect of baicalin combined with azithromycin and quinolone antibiotics on biofilms of multidrug-resistant Pseudomonas aeruginosa.Methods Ten strains of multidrug-resistant Pseudomonas aeruginosa were randomly selected.The minimum inhibitory concentrations(MICs)of baicalin,levofloxacin,ciprofloxacin,and azithromycin alone and in combination were determined using the microbroth dilution method,and the fractional inhibitory concentration index(FICI)was calculated to assess the effect of combined drug therapy.Additionally,the crystal violet staining method was used to evaluate the inhibitory effect of the drugs on biofilms.Results Baicalin showed synergistic or additive effects when combined with levofloxacin and ciprofloxacin.The combination of quinolone antibiotics with baicalin exhibited inhibitory effects on the biofilms of all ten Pseudomonas aeruginosa strains;the inhibitory effect of azithromycin combined with low concentrations of levofloxacin and ciprofloxacin on biofilms showed statistically significant differences(P<0.05).Conclusion The effect of combined Chinese and Western medication is superior to monotherapy.Levofloxacin and ciprofloxacin can inhibit the growth of Pseudomonas aeruginosa biofilms,and their inhibitory effects on biofilms when combined with baicalin show no significant difference.Azithromycin can enhance the efficacy of low-concentration quinolone antibiotics.
