蒲公英花色素提取工艺优化及其稳定性研究

Optimization of extraction process of dandelion flower pigment and its stability

摘要 以蒲公英花为原料,采用超声波辅助甲醇提取法进行蒲公英花色素的提取,通过单因素和响应面试验对提取工艺进行优化,同时对蒲公英花色素稳定性进行考察。试验结果显示,蒲公英花色素的最佳制备工艺条件为料液比1∶22.8 g/mL、提取时间9.9 min和提取温度45.8℃,在此工艺条件下,蒲公英花色素提取物平均吸光度值为1.565±0.06,与模型预测值误差为0.64%,证明本试验响应面模型所得优化工艺有效可行。稳定性试验结果显示,蒲公英花色素在20~50℃之间有着较好的热稳定性,同时不能长时间接触日光直接照射;该色素在pH值2~8的环境下应用稳定性较好,但不适宜在强碱性环境下应用;离子试验结果显示,锌离子对蒲公英花色素影响最显著。该研究结果可为蒲公英花色素的开发应用奠定理论基础。 Dandelion flowers were used as raw materials,dandelion flower pigment was extracted by ultrasonicassisted methanol extraction and the extraction process was optimized by single factor and response surface methodology.Pigment stability under various conditions was also investigated.The optimum extraction was achieved with a solid-to-liquid ratio of 1∶22.8 g/mL,an extraction time of 9.9 min and an extraction temperature of 45.8 ℃.Under these conditions,the average absorbance value of the pigment was 1.565±0.06,with a deviation of 0.64% from the predicted model value,indicating that the optimized process condition obtained was effective and feasible.Stability test revealed that the pigment exhibited good thermal stability between 20~50 ℃,but prolonged exposure to direct sunlight was not recommended.The pigment remained stable within a pH range of 2~8,but was unstable in strong alkaline environments.Ion tests indicated that zinc had the most significant impact on dandelion flower pigment stability.These findings provide a valuable foundation for the development and application of dandelion flower pigment.
