HPLC法同时测定引种紫锥菊中3种酚酸类成分

摘要 试验旨在建立紫锥菊中绿原酸、菊苣酸、咖啡酸同时测定的HPLC分析方法。试验对紫锥菊中3种酚酸类成分的测定方法进行参数优化,色谱条件为Agilent (TC-C_(18),4.6 mm×250 mm,5μm)色谱柱,流动相乙腈-0.1%磷酸,梯度洗脱,流速1.0 mL/min,检测波长330 nm,柱温30℃,比较蒙自市引种栽培紫锥菊根、茎、叶、头状花序和花序梗中3种酚酸类成分的含量。结果显示,3种酚酸类成分在色谱条件下线性关系、精密度、稳定性、重复性和加样回收率良好。经过系统的方法学考察,该法定性定量准确、可靠,可用于紫锥菊药材及制剂的质量评价。研究表明,蒙自市引种栽培生产的紫锥菊药材中3种酚酸类成分含量较高,可作为优质饲料添加剂和中兽药原料药材生产基地。 The experiment aimed to establish an HPLC analytical method for the simultaneous determination of chlorogenic acid,cichoric acid,and caffeic acid in Echinacea purpurea L..The experimental method for the determination of the three phenolic acids in Echinacea purpurea L.was optimized.The chromatographic conditions were as follows:Agilent(TC-C_(18),4.6 mm×250 mm,5 μm) column,mobile phase of acetonitrile-0.1% phosphoric acid,gradient elution,flow rate of 1.0 mL/min,detection wavelength of 330 nm,and column temperature of 30 ℃.The contents of the three phenolic acids in the roots,stems,leaves,capitulum,and peduncle of Echinacea purpurea L.introduced and cultivated in Mengzi city were compared.The results showed that the three phenolic acid components had good linearity,precision,stability,repeatability,and spiked recovery rate under chromatographic conditions.After systematic methodological examination,the method was found to be accurate and reliable for the qualitative and quantitative analysis of Echinacea purpurea L.medicinal materials and preparations.The study indicates that the Echinacea purpurea L.cultivated in Mengzi City has a high content of the three phenolic acid components,making it a potential base for high-quality feed additives and raw materials for traditional Chinese veterinary medicine.
