摘要
目的 研究健脾复方胃肠安抑制胃癌细胞侵袭迁移的机制。方法 CCK8检测胃肠安对胃癌MGC803细胞增殖的影响。采用细胞侵袭迁移实验检测胃肠安和转化生长因子β1(TGF-β1)干预后MGC803细胞迁移和侵袭能力的变化。Western blot检测胃肠安和TGF-β1干预后各组细胞EMT相关蛋白表达情况。荧光双染法检测RUFY3和F-actin表达情况。结果 胃肠安抑制MGC803细胞的增殖呈时间浓度依赖性,48 h和72 h的半数抑制浓度(IC50值)分别为856、781 mg·L^(-1)。TGF-β1可以诱导胃癌MGC803细胞EMT的发生。胃肠安能够逆转TGF-β1对MGC803细胞运动的促进作用。胃肠安抑制了EMT模型中侵袭迁移和MMP家族相关蛋白的表达,并且胃肠安抑制MGC803细胞侵袭力与降低RUFY3的表达相关。结论 胃肠安能够通过RUFY3抑制胃癌MGC803细胞侵袭迁移。
Objective To study the mechanism of spleen-strengthening traditional Chinese medicine(TCM)prescriptions Weichang'an(胃肠安)inhibiting the invasion and migration of gastric cancer cells.Methods CCK8 was used to detect the effect of Weichang'an on the proliferation of gastric cancer MGC803 cells.Cell invasion and migration experiment was used to detect the changes of migration and invasion ability of MGC803 cells after the intervention of Weichang'an and transforming growth factor-β1(TGF-β1).The expression of EMT-related proteins in each group was detected through Western blot.Phalloidin staining was used to detect the expressions of RUN and FYVE domain protein 3(RUFY3)and F-actin.Results The proliferation of MGC803 cells was time-dependent with the IC50 of 856 and 781 mg·L^(-1) at 48 h and 72 h,respectively.TGF-a1 can induce EMT in gastric cancer MGC803 cells.Weichang'an reversed the promoting effect of TGF-a1 on the motility of MGC803 cells.The invasion and migration of MGC803 cells and the expression of MMP family related proteins were inhibited in EMT model,and the inhibition of invasion of MGC803 cells was correlated with the decrease of RUFY3 expression.Conclusion The invasion and migration of MGC803 cells in gastric cancer was inhibited by RUFY3.
