摘要
目的 探讨镉对小鼠初级精母(GC-2spd)细胞G1期和细胞周期蛋白Cyclin D1和Cyclin E1的影响。方法 将处于对数生长期的GC-2spd细胞暴露于终浓度为0(对照)、2.5、5、10μmol/L的氯化镉培养液染毒24 h。采用流式细胞技术测定细胞周期变化,采用Western blot法检测细胞周期蛋白Cyclin D1和Cyclin E1的表达。结果 与对照组相比,仅10μmol/L氯化镉染毒组GC-2spd细胞G1期细胞的构成比较高,差异有统计学意义(P<0.01);且随着氯化镉染毒浓度的升高,GC-2spd细胞G1期细胞的构成比呈上升趋势。说明镉染毒可使GC-2spd细胞周期阻滞于G0/G1期。与对照组相比,10μmol/L氯化镉染毒组GC-2spd细胞Cyclin D1蛋白的表达水平以及5、10μmol/L氯化镉染毒组Cyclin E1蛋白的表达水平下调,差异均有统计学意义(P<0.05)。结论 镉可使生精细胞周期蛋白Cyclin D1和Cyclin E1表达下调,从而导致GC-2spd细胞G1期发生阻滞,最终抑制细胞增殖。
Objective To understand the effect of cadmium on the G1 phase of cell cycle and the expression of cyclins including Cyclin D1 and Cyclin E1 in GC-2spd cells.Methods GC-2spd cells were divided into groups and exposed to the cadmium chloride at 0(the control group),2.5,5 and 10μmol/L respectively for 24 hours.Cell cycle changes were measured by flow cytometry,and the expression of cyclin D1 and cyclin E1 were detected by Western blot.Results Compared with the control group,the proportion of G1-phase cells in the 10μmol/L cadmium chloride group significantly increased(P<0.01,and the G1 phase of cell cycle in GC-2spd cells concentration presented an increasing trend with the increase of cadmium chloride exposure doses,and this showed that cadmium exposure could block the GC-2spd cell cycle in the G0/G1 phase.Compared with the control group,the protein expression of Cyclin D1 in the 10μmol/L exposure group decreased significantly,and the protein expression of Cyclin E1 in the 5μmol/L and 10μmol/L exposure group significantly decreased(P<0.05).Conclusion Cadmium can down-regulate the expression of spermatogenic cyclins Cyclin D1 and Cyclin E1,arrested of G1 phase of GC-2spd cells and ultimately inhibits cell proliferation.
