摘要
目的 提供改良的制作大鼠局部缺血性脑损伤模型的方法。方法 雄性Wistar大鼠 44只 ,体重 1 60~1 80g,随机分为空白对照组 (n =6)、假手术组 (n =1 5)、缺血损伤再灌注组 (n =2 3) ,35只用于脑的大体和组织学研究 ,9只用于核磁共振T2 WI成像研究。颈部正中切口 ,采用从颈内动脉插入线栓的造模方法 ,用 5/ 0尼龙线栓造成大脑中动脉血供阻断 (MCAO) 2h。缺血损伤再灌注组的再灌时间点为 1d(n =8)、2d(n=5)、3d(n =5)、7d(n =5)。采用大体和组织学观察方法 ,研究脑组织梗死灶及血管的变化。采用核磁共振T2 WI成像技术 ,检测大脑高信号强度区信号的变化。结果 空白对照组、假手术组未见梗死灶 ,缺血损伤再灌注组有梗死灶的形成 ,7d时的梗死灶体积为 (1 1 8.65± 43 .61 )mm3。结论 改良的线栓阻断大脑中动脉制作缺血性脑损伤模型的方法稳定可靠 ,手术过程简化 ,容易操作。
Objective To develop a simple, relatively noninvasive model of focal ischemic brain injury in rats. Methods Fourty four male Wistar rats with an average body weight of 170±10 g (mean±SD) were used, and were randomly divided into three groups: group A rats ( n =6) underwent no operation,group B rats ( n =15) underwent sham operation,group C rats ( n =23) underwent an operation of ischemic brain injury,in which the occlusion of right middle cerebral artery was induced by 5/0 nylon suture for 2 hours.The time points of reperfusion for group C rats were 1 day ( n =8), 2 days ( n =5),3 days ( n =5) and 7 days ( n =5).The infarct volume and vascular changes in brains of group C rats were observed by gross anatomy and histology,and the changes of high signal intensity zones were measured by nuclear magnetic resonance T 2 weighted images. Results No infarct zone was found in group A and group B rats, the infarct volume at 7 days after reperfusion in group C rats was 118.65± 43.61 mm 3 . Conclusion The reported modified suture occlusion method for preparation of model of ischemic brain injury is reliable, and easier for management.
