亚低温对大鼠脑缺血再灌注损伤后Bcl-2、Bax和C-fos蛋白表达及损伤神经细胞凋亡的影响 被引量:6

Effects of mild hypothermia on Bcl-2,Bax and C-fos protein expression and cell apoptosis after cerebral ischemia/reperfusion injury in rats

摘要 目的 观察亚低温对大鼠脑缺血再灌注损伤后Bcl-2、Bax和C fos蛋白表达及损伤神经细胞凋亡的影响。方法 采用大鼠局灶性脑缺血再灌注损伤模型 ,大脑中动脉阻塞 2h ,再灌注损伤 6h ,用原位缺口末端标记 (TdtmediateddUTP -biotinnickendlabeling ,TUNEL)法和免疫组织化学法分别检测假手术组、对照组和亚低温组的凋亡细胞百分率和Bcl-2、Bax和C -fos蛋白表达水平。结果 亚低温组Bcl -2蛋白表达水平为 (63 .48± 0 .47) % ,较对照组显著升高 (P <0 .0 5 ) ,而Bax和C -fos蛋白表达水平和凋亡细胞百分率分别为 (6.13± 0 .93 ) %和 (5 .0 9± 0 .2 1) % ,明显低于对照组 (P <0 .0 5 )。结论 亚低温下调大鼠脑缺血再灌注损伤后Bax和C -fos蛋白表达水平和上调Bcl -2蛋白表达水平 ,可能与其抗损伤神经细胞凋亡作用有关。 Objective To observe the effects of mild hypothermia on Bcl-2,Bax and C-fos protein expression and apoptosis of the injuried nervous cell after focal cerebral ischemia/reperfusion injury in rats. Methods Models of focal cerebral ischemia/reperfusion injury were prepared by 2 hours of occlusion followed by 6 hours of reperfusion of the middle cerebral arteries(MCA) in rats. TUNEL and immunohistochemical technique were used to examine the apoptotic cell rate and the level of Bcl-2,Bax and C-fos protein expression in the sham-operated group, the control group and the mild hypothermia group, respectively. Results The level of Bcl-2 protein expression(63.48± 0.47 )% was significantly higher in the mild hypothermia group than that in the control group( P < 0.05 ). The level of Bax and C-fos protein expression(6.13±0.93%,5.09±0.21%) and the apoptotic cell rate were significantly lower in the mild hypothermia group than those in the control group( P <0.05). Conclusion The decreased level of Bax and C-fos protein expression and the increased level of Bcl-2 protein expression in rats of hypothermia group may be closely related to the decreased rate of cell apoptosis after cerebral ischemia/reperfusion injury.
