摘要
目的 建立敏感性高、特异性强 ,方法简便和经济实用的快速诊断囊尾蚴病的免疫学检测方法。方法将猪囊尾蚴液抗原点加于微孔滤膜 ,经预作用和封闭后 ,依次加入待检血清和金标记二抗 ,建立快速斑点免疫金染色法 (F -Dot-IGS)以检测囊尾蚴病患者血清抗体 ,同时用快速微量斑点免疫金银染色法 (RMDot-IGSS)和斑点免疫金银染色法 (Dot-IGSS)检测作为对照。结果 检测 4 0例囊尾蚴病患者血清抗体 ,阳性率 10 0 % ;检测 4 0例健康人血清均为阴性 ;除包虫病患者外 ,10例肝吸虫病患者、10例血吸虫病患者和 10例肺吸虫病患者血清未见交叉反应。结论 F -Dot-IGS敏感特异 ,快速简便 ,有较高的实用价值 。
Objective To set up a specific, sensitive, convenient, economical and fast immunological assay for detection of antibodies in the patients with cysticercosis.?Methods The fast-dot-immunogold staining assay (F-Dot-IGS) was performed as follows: The cysticercus cyst fluid antigens of Taenia Solium were added onto micropore filter membrane. After pre-reacting and blocking, the sample serum to be tested and the sheep anti-human IgG labeled with chloroauric acid were added sequentially to read the resault. As controls, the dot immunogold silver staining (Dot-IGSS) and rapid microvolume Dot-IGSS (RM Dot-IGSS) were performed simultaneously.?Results All of the 40 confirmed cases of cysticercosis showed positive reaction to the F-Dot-IGS. The 40 healthy controls, 10 cases of clonorchiasis, 10 schistosomiasis and 10 paragonimiasis, all showed negative reactions, except for the 10 cases of echinococcosis, in which obvious cross reaction occurred.?Conclusion Compared with Dot-IGSS and RM Dot-IGSS, F-Dot-IGS has similar sensitivity and specificity, is more rapid, simpler and more economical, and hence is worth application in field situations and for rapid clinical diagnosis.
