薰鲁香中1个新的降三萜类化合物 被引量:4

摘要 综合运用多种现代色谱分离方法从薰鲁香中分离得到10个化合物,通过HR-ESI-MS、IR、UV、NMR、量子化学计算核磁共振参数(qcc-NMR)等技术和文献对比,分别鉴定为17β-hydroxy-28-norolean-18-en-3-one(1)、28-norolean-12,17-dien-3,11-dione(2)、28-norolean-16,18-dien-3-one(3)、(24Z)-26-hydroxy-7,24-dientirucalla-3-one(4)、masticadienonic acid(5)、mastica-dienolic acid(6)、erythrodiol(7)、3β,28-dihydroxyoleana-11,13(18)-diene(8)、3-oxo-olean-9(11),12-dien-28-oic acid(9)、12-oleane-3,11-dione(10)。其中,化合物1为新化合物,化合物2为新天然产物,化合物1~2、4、7~9均为首次从薰鲁香中分离得到。化合物1可显著抑制脂多糖(LPS)诱导的小鼠单核巨噬细胞RAW264.7中NO的产生,IC_(50)为(7.44±0.49)μmol·L^(-1),强于阳性对照地塞米松[dexamethasone, IC_(50)为(9.93±1.17)μmol·L^(-1)],化合物2~3、5、9~10显示出一定的抑制效果,IC_(50)在14.82~28.82μmol·L^(-1);化合物4~5、7可显著抑制人结肠腺癌细胞SW480的生长,IC_(50)分别为(16.13±1.69)、(27.40±1.81)、(10.01±0.10)μmol·L^(-1)。 One new triterpenoid 17β-hydroxy-28-norolean-18-en-3-one(1)and three compounds with first reports of 2D NMR data 28-norolean-12,17-dien-3,11-dione(2),28-norolean-16,18-dien-3-one(3),(24Z)-26-hydroxy-7,24-dientirucalla-3-one(4),along with six known ones(5-10),were isolated from mastic by using various modern chromatographic separation methods.Their structures were determined based on HR-ESI-MS,IR,UV,NMR,quantum chemical calculation of NMR(qcc-NMR)and comparison with those reported in literature.Anti-inflammatory experiments showed that compound 1 significantly inhibited LPS-induced NO production in mouse monocyte macrophage RAW264.7 cells with an IC_(50) of(7.44±0.49)μmol·L^(-1),which was more potent than the positive control,dexamethasone[IC_(50)=(9.93±1.17)μmol·L^(-1)];compounds 2-3,5,9-10 showed some inhibitory effects,with IC_(50) values ranging from 14.82 to 28.82μmol·L^(-1).The anticancer experiments showed that compounds 4-5 and 7 could markedly inhibit the growth of human colon adenocarcinoma cells SW480 with IC_(50) values of(16.13±1.69),(27.40±1.81),and(10.01±0.10)μmol·L^(-1),respectively.
