摘要
目的建立油菜花粉颗粒质量标准。方法采用薄层色谱法(thin-layer chromatography,TLC)对油菜花粉颗粒中黄酮类成分进行定性鉴别;运用高效液相色谱法(high performance liquid chromatography,HPLC)对槲皮素、柚皮素、山柰酚和异鼠李素(简称槲皮素)等4指标成分进行定量分析。结果槲皮素、山柰酚薄层色谱斑点清晰,分离良好;槲皮素等4指标成分在浓度为10~160 mg/L(r=0.9999)、4~64 mg/L(r=0.9993)、30~480 mg/L(r=0.9999)、0.5~8 mg/L(r=0.9993)时线性关系良好;平均回收率与相对标准偏差(relative standard deviation,RSD)分别为95.9%(1.50%)、102.7%(3.62%)、98.4%(1.36%)和98.8%(4.44%)。结论HPLC操作简便、准确可靠、专属性强,可用于油菜花粉颗粒的质量控制。
Objective:To establish the quality standard of rape pollen granules.Methods:The flavonoids in rape pollen granules were qualitatively identified by TLC.Quercetin,nsringin,kaemoferol and isorhamnetin(Quercetin and other index components)were quantitatively analyzed by HPLC.Results:The spots of quercetin and kaempferol were clear and well separated by TLC.Quercetin and other index components showed a good linear relationship in the range of 20~320 mg/L(r=0.9999),4~64 mg/L(r=0.9993),60~960 mg/L(r=0.9999),0.5~8 mg/L(r=0.9993).The average recovery(RSD)were 96.7%(2.83%),102.7%(3.61%),98.2%(1.29%),and 98.9(4.43%).Conclusion:The method is simple,reliable and specific,which can be used for the quality control of rape pollen granules.
