摘要
为了探讨人白细胞介素 18(hIL 18)的表达对鼻咽癌细胞体内致瘤性的影响 ,我们构建了hIL 18的真核表达载体pcD NA3/hIL 18,转染人鼻咽癌细胞株SUNE细胞 ;然后以转染阳性的SUNE细胞和未转染的SUNE细胞接种到用健康人外周血白细胞免疫重建的SCID鼠 (hu PBL SCID鼠 ) ,观察成瘤情况。结果发现 ,所构建的真核表达载体在鼻咽癌细胞中能高效表达hIL 18;ELISA法测得转染阳性细胞培养上清液中hIL 18的含量为 (85± 10 )pg/ml,而未转染的SUNE细胞的培养上清液中hIL 18的含量低于 5pg/ml。将转染阳性的细胞克隆和未转染的SUNE细胞接种hu PBL SCID小鼠后连续观察 5 0d ,发现前者形成的肿瘤其生长速度明显慢于后者 ,其平均瘤重也有显著性差异 (P <0 0 0 1)。说明在体内hIL 18的表达能有效地抑制鼻咽癌细胞的成瘤作用。
To study the tumorigenicity of nasophary ngeal carcinoma (NPC) cells in the hu-PBL-SCID mouse model,human IL-18 cDNA had been amplified from PBMC through RT-PCR,cloned and then subcloned into eukaryotic expression vector pcDNA3 at EcoR I and BamH I si tes and transfected into a NPC-derived cell-line SUNE cells The transfected and non-transfected SUNE cells were inoculated subcutaneously in the right flan k of hu-PBL-SCID mice. Results showed that the recombinant eukaryotic expressi on vector could highly express the hu-IL-18 in SUNE cells but no hu-IL-18 ex pression in the control cells The concentration of hu-IL-18 in the supernata nts of the transfected SUNE cell cultures as detected by ELISA was (85±10)pg/ml but that in the supernatants of non-transfected cells was less than 5 pg/ml After inoculation to the hu-PBL-SCID mice,the pcDNA3/hu-IL-18 trans fected SUNE cells grew slowly than the non-transfected cells The tumors were excised 50 days later and weighed,and it was found by statistical analysis that the size and mean weight of the tumors from the former group of mice were signif icantly lower in contrast to those of the latter These results indicate that t he tumorigenicity of nasopharyngeal carcinoma cells could be effectively weaken by the expression of IL-18 in vivo -
