高丹草/黄芪间作对高丹草根际土壤微生物多样性的影响 被引量:7

Effects of Sorghum-Sudangrass Hybrid and Astragalus membranaceus Intercropping on Rhizosphere Soil Microbial Community Diversity

摘要 为探究高丹草/黄芪间作对高丹草根际土壤微生物多样性的影响,以高丹草单作为对照(S),设置3种高丹草与黄芪的间作模式(2∶1,2∶2,4∶2),通过测定高丹草农艺性状以及采用Illumina PE250高通量测序,分析高丹草/黄芪间作对高丹草生物性状和高丹草根际土壤微生物多样性的影响,结果表明,不同时期不同高丹草间作模式下高丹草株高、茎粗、单株鲜重和单株干重等生物性状总体上高于高丹草单作模式,且就平均而言高丹草/黄芪2∶1的间作模式效果最好。采用Illumina PE250高通量测序技术对高丹草根际土壤细菌16S rDNA和真菌ITS rDNA进行测定,结果表明,通过对4个样品土壤细菌16S测序共得到优化序列172643条,碱基对数范围为13515561~22878067 bp,平均序列长度分布范围为415.62~417.42 bp。高丹草/黄芪间作使高丹草根际微生物群落Chao 1指数和Ace指数均显著大于单作模式(P<0.05),高丹草根际土壤的优势菌种(相对丰度大于10%)分别为变形菌门、放线菌门、酸杆菌门和绿弯菌门,3种不同间作模式土壤变形菌门的相对丰度高于高丹草单作土壤,而酸杆菌门的相对丰度低于高丹草单作土壤。不同高丹草间作模式下,土壤的微生物群落组成发生了变化,说明高丹草/黄芪间作模式可以改善高丹草根际土壤细菌和真菌微生物丰度和多样性,为推广应用高丹草多样性种植方式提供一定的理论依据。 This study aimed to explore the effects of sorghum-sudangrass hybrid and Astragalus membra⁃naceus intercropping on agronomic traits and rhizosphere soil microbial diversity in sorghum-sudangrass hybrid using Illumina PE250 high-throughput sequencing.Three modes of sorghum-sudangrass hybrid and Astragalus membranaceus intercropping(2∶1,2∶2,4∶2)were set up in this experiment,with sorghumsudangrass hybrid monocropping as control(S).The results showed that the agronomic traits of sorghumsudangrass hybrid,such as plant height,stem diameter,fresh weight per plant,and dry weight per plant were generally higher under different intercropping modes of different periods compared to monocropping.The I1 treatment exhibited the optimum performance.Four samples of soil bacteria were subjected to 16S sequencing,which obtained a total of 172643 optimized sequences.The number of base pairs ranged from 13515561 bp-22878067 bp,and the average sequence length distribution ranged from 415.62 bp to 417.42 bp.The Chao1 and Ace indexes in the intercropping treatments were significantly higher than those in the control group(P<0.05).The dominant strains in the soil were Proteobacteria,Actinobacteria,Acidobacteria,and Chloroflexi,with relative abundances greater than 10%.Among the dominant strains,the relative abundance of Proteobacteria in intercropping treatments was higher than that in the control group,with the highest relative abundance of Acidobacteria in the control group.The composition of the soil microbial com‑munity changed under different intercropping modes,suggesting that sorghum-sudangrass hybrid and Astragalus membranaceus intercropping could improve the abundance and diversity of bacteria and fungi in the rhizosphere soil.These findings provide a theoretical basis for the popularization of the diverse planting methods of sorghum-sudangrass hybrid.
