Study on the Metabolites Analysis of Different Strains of Mesona chinensis Benth.Based on UPLC-QTOF-MS

摘要 目的基于代谢组学方法探究凉粉草不同栽培品系(台湾草、平远草和增城草)的差异代谢物,筛选品系鉴别的主要代谢物。方法运用超高效液相色谱-四极杆飞行时间质谱(UPLC-Q-TOF-MS)非靶向代谢组学技术分析不同品系凉粉草的化学成分,数据经主成分分析、正交偏最小二乘判别分析和方差分析处理,筛选不同品系鉴别的代谢物。结果共筛选得到186种差异代谢物,台湾草、平远草与增城草之间的差异代谢物主要显著富集于黄酮类生物合成通路。3个品系均有芸香苷、山奈酚3-O-葡萄糖苷、槲皮素等黄酮类代谢物;结合差异倍数和文献报道,槲皮素、山奈酚3-O-葡萄糖苷可作为3个凉粉草品系区分的差异代谢物。结论UPLC-Q-TOF-MS代谢组学技术可阐明不同品系凉粉草化学成分差异,为其品系鉴别和资源开发提供参考。 Objective To explore the differential metabolites in different cultivated strains of Mesona chinensis Benth.(Taiwan grass,Pingyuan grass and Zengcheng grass)by metabolomics technique,and to screen main metabolites for strain identification.Methods Non-targeted UPLC-Q-TOF-MS-based metabolomics technology was used to analyze the chemical composition of dfferent strains of Mesona chinensis Benth.,and the data were processed by principal component analysis,orthogonal partial least squares discriminant analysis and ANOVA to screen metabolites for strain identification.Results A total of 186 metabolites were screened,and the differential metabolites of Taiwan grass,Pingyuan grass,and Zengcheng grass were significantly enriched in flavonoid biosynthetic pathway.There were flavonoid metabolites in three strains,such as rutin,kaempferol-3-O-glucoside,quercetin,etc.Combining the differential multiples with literature reports,we selected quercetin and kaempferol-3-O-glucoside as differential metabolites for distinguishing three strains of Mesona chinensis Benth..Conclusion The UPLC-Q-TOF-MS metabolomics is able to clarify the differences in chemical composition of different strains of Mesona chinensis Benth.and provide reference for its strain identification as well as for resource development.
