摘要
目的探索ZNF165在食管癌中的表达,分析ZNF165表达与食管癌患者临床病理学特征和预后的相关性。方法通过TCGA和GEO数据库分析ZNF165在食管癌中的表达情况。利用Logistic回归分析ZNF165表达与食管癌患者临床病理特征的相关性。接着利用Kaplan-Meier分析、ROC曲线、Cox回归分析ZNF165在食管癌患者中的预后价值。通过TCGA数据库寻找ZNF165共表达基因,进行GO生物功能及KEGG通路富集分析。通过肿瘤免疫评估资源(Tumor Immune Evaluation Resource,TIMER)数据库分析ZNF165与食管癌免疫浸润的相关性。最后,通过qRT-PCR验证ZNF165在人食管癌细胞系和食管癌组织标本中的表达。结果TCGA和GEO数据集分析表明,食管癌组织样本中ZNF165表达显著高于非肿瘤组织(P<0.05),qRT-PCR进一步证实了这一点。高表达ZNF165的食管癌患者总生存期明显短于ZNF165低表达患者(P=0.024)。Cox回归分析提示,ZNF165是食管癌患者总生存期的独立预测指标(P=0.005)。基于TNM分期、组织学分级、年龄和ZNF165表达的预测模型能有效预测食管癌患者1年、3年及5年总生存率。GO生物功能及KEGG通路富集分析提示,ZNF165主要参与抗菌体液反应、脂肪消化和吸收、溶质-钠离子共转运活性等生物学过程。免疫相关性分析提示,ZNF165表达与嗜酸性粒细胞、NK-CD56 bright、Th17等24个免疫细胞具有相关性,与多种免疫细胞表面标志物显著相关。结论ZNF165表达与食管癌患者预后及免疫浸润具有相关性,有可能作为食管癌诊断及预后预测的重要生物学靶点。
Objective To explore the expression of ZNF165 in esophageal cancer and to analyze the correlation between ZNF165 expression and clinicopathological features and prognosis of esophageal cancer patients.[WTHZ]Methods The expression level of ZNF165 in esophageal cancer was analyzed by TCGA and GEO databases.Logistic regression was used to analyze the correlation between ZNF165 expression and clinicopathological features of esophageal cancer.Kaplan-Meier,ROC curve,Cox regression were then used to evaluate the prognostic value of ZNF165 in patients with esophageal cancer.ZNF165 co-expressed gene was searched through TCGA database.GO biological function and KEGG pathway enrichment analysis were performed.The association between ZNF165 and immune invasion in esophageal cancer was analyzed by the Tumor Immune Evaluation Resource(TIMER)database.Finally,the expression of ZNF165 in human esophageal cancer cell lines and esophageal cancer tissue specimens was verified by qRT-PCR.[WTHZ]Results TCGA and GEO dataset analysis showed that ZNF165 expression was significantly higher in esophageal cancer tissue samples than in non-tumor tissues,which was further confirmed by qRT-PCR(P<0.05).The overall survival of patients with esophageal cancer with high expression of ZNF165 was significantly lower than that of patients with low expression of ZNF165(P=0.024).Cox regression analysis showed that ZNF165 was an independent predictor of overall survival of esophageal cancer patients(P=0.005).Prediction models based on TNM stage,histological grade,age and ZNF165 expression could effectively predict the 1-year,3-year and 5-year overall survival rates of esophageal cancer patients.GO biological function and KEGG pathway enrichment analysis showed that ZNF165 was mainly involved in biological processes such as antibacterial humoral reaction,fat digestion and absorption,and solute-sodium ion co-transport activity.Immunocorrelation analysis showed that ZNF165 expression was correlated with 24 immune cells such as eosinophils,NK-CD56 bright and Th17,and was significantly associated with a variety of immune cell surface markers.[WTHZ]Conclusion The expression of ZNF165 is related to the prognosis and immune invasion of patients with esophageal cancer,and may be used as an important biological target for the diagnosis and prognosis of esophageal cancer.
