摘要
目的:观察补肾活血祛风复方对阿霉素肾病大鼠肾组织中NOD样受体蛋白3(NLRP3)表达的影响,探讨补肾活血祛风复方治疗肾病综合征的作用机制。方法:60只SD雄性大鼠随机分为5组:空白组、模型组、泼尼松组、补肾活血祛风复方组和补肾活血祛风复方+泼尼松组。空白组和模型组予蒸馏水灌胃;泼尼松组予醋酸泼尼松片混悬液灌胃;补肾活血祛风复方组给予补肾活血祛风复方药液灌胃;补肾活血祛风复方+泼尼松组上午以醋酸泼尼松片混悬液灌胃,下午以补肾活血祛风复方药液灌胃;连续灌胃6周。检测24小时尿蛋白定量、肾组织的NLRP3平均光密度及mRNA表达含量。结果:24小时尿蛋白定量:泼尼松组、补肾活血祛风复方+泼尼松组与模型组相比大鼠尿蛋白有所降低(P<0.05)。各组大鼠肾组织NLRP3表达的改变:模型组NLRP3mRNA及NLRP3蛋白表达较空白组升高(P<0.05),各治疗组NLRP3mRNA及NLRP3蛋白表达比模型组降低(P<0.05),补肾活血祛风复方+泼尼松组NLRP3mRNA表达显著降低(P<0.01)。结论:补肾活血祛风复方组+泼尼松在降低尿蛋白及NLRP3表达最为显著。
Objective:To observe the effect of Bushen Huoxue Qufeng decoction on Nod-like receptor protein 3(NLRP3)expression in kidney tissue of adriamycin nephropathy rats,and to explore the mechanism of Bushen Huoxue Qufeng decoction in the treatment of nephrotic syndrome.Methods Sixty male SD rats were randomly divided into 5groups:blank group,model group,prednisone group,Bushen Huoxue Qufeng decoction group and Bushen Huoxue Qufeng decoction+prednisone group.Blank group and model group were given distilled water intragastric administration.Prednisone group was given prednisone acetate tablet sus-pension by gavage.Bushen Huoxue Qufeng decoction group was given decoction liquid by gavage;Bushen Huoxue Qufeng decoc-tion+Prednisone group was given prednisone acetate tablet suspension in the morning and decoction in the afternoon.Continuous gavage for 6weeks.The 24H urinary protein,the mean optical density of NLRP3and mRNA expression in renal tissue were meas-ured.The mean NLRP3optical density of renal tissue was determined by immunohistochemical method.The mRNA expression of NLRP3was detected by Real-time PCR.Results:24H urine protein quantification:Compared with model group,prednisone group,Bushen Huoxue Qufeng decoction+Prednisone group decreased urine protein(P<O.05).Changes of NLRP3expression in renal tissue of rats in each group:NLRP3mRNA and NLRP3protein expressions in model group were increased compared with blank group(P<0.05),and NLRP3mRNA and NLRP3protein expressions in treatment groups were decreased compared with model group(P<0.05).NLRP3mRNA expression in Bushen Huoxue Qufeng decoction+Prednesone group was significantly de-creased(P<0.01).Conclusion:Bushen Huoxue Qufeng decoction+prednisone significantly decreased the expression of urinary protein and NLRP3.
