肾康灵干预阿霉素肾病大鼠NF-kB的实验研究 被引量:17

摘要 观察益肾活血中药肾康灵对阿霉素肾病大鼠(AN)的干预作用及与核因子(NF kB)的关系,探讨小儿原发性肾病综合征(PNS)的发病机理及益肾活血中药干预治疗的可能机制。用阿霉素(ADR)诱导大鼠类似于人类微小病变型肾病(MCD)模型,用强的松加中药肾康灵治疗,并设对照组。结果:AN鼠治疗前NF kB活性显著增强(P <0 0 1) ,尿蛋白显著增多,Alb显著下降,Ch和TG显著升高,肾脏病理损害明显加重;治疗组AN鼠在中药肾康灵加强的松干预下,上述指标都明显改善(P <0 0 5或P <0 0 1) ,肾脏病理损害明显减轻。结论:益肾活血中药肾康灵加强的松能显著改善AN鼠的肾虚血瘀证表现,降低NF kB活性。NF kB可作为中医肾虚血瘀证诊断和病情预后转归判断的客观指标。 To observe the curative effects of Shenkangling, the function of benefiting kidney and promoting blood circulation, on the adriamycin-induced nephrotic(AN)rats, and its relationship with nuclear factor-Kappa B(NF-kB) in AN rats, then to explain the pathogenesis of primary nephrotic syndrome(PNS) and the mechanisms of Chinese medication of benefiting kidney and promoting blood circulation. The adriamycin-induced rat models, which are similar to human minimal change nephropathy (MCD), were treated by prednisone with Shenkangling. Meanwhile, control groups were established. Result: NF-kB enhanced evidently( P 〈 0.01 ), the urine protein, CH and TG increased clearly, Alb decreased markedly, and the renal pathomorphology aggravated in AN rats. All the above-mentioned indexes ameliorated obviously (P 〈 0.05 or P 〈 0.01 ), and the renal lesion markedly lessened after treatment. Conclusions Shenkangling with prednisone can remarkably ameliorate the clinical situation of the symptom of kidney deficency and blood stasis, as well as reduce the increased activity of NF-kB in AN rats, and NF-kB can be the index of the diagnosis and prognosis of symptom of kidney deficiency and blood stasis.
