摘要
【目的】探究盐驯化对青花菜组培苗耐盐性的影响。【方法】本试验以青花菜组培苗为材料,使用含75、100、150、200 mmol/L NaCl的液体培养基对青花菜组培苗驯化7 d,分析高盐胁迫(225 mmol/L NaCl)7 d,青花菜组培苗的生理指标、硫代葡萄糖苷含量的变化,并对其生理指标进行隶属函数分析,探究青花菜组培苗对盐驯化响应的生理机制。【结果】青花菜组培苗的各项生理指标对盐驯化有不同程度的响应。盐驯化使青花菜组培苗的POD活性,脯氨酸、可溶性糖和硫代葡萄糖苷含量较CK2(盐对照)显著增加,MDA含量较CK1(空白对照)、CK2显著降低,可溶性蛋白含量与CK2无显著差异,总酚、类黄酮含量较CK2显著降低。SOD活性在150 mmol/L NaCl驯化组中显著高于CK2,与CK1同水平,花青素含量在200 mmol/L NaCl驯化组显著高于CK1、CK2。隶属函数分析得出,150 mmol/L NaCl驯化组在高盐胁迫下各项生理指标综合得分最高。【结论】盐驯化可以提高青花菜组培苗的耐盐性,以150 mmol/L NaCl驯化效果最好。
【Objective】It is to investigate the effect of salt acclimation on the salt tolerance of broccoli tissue cultured seedlings.【Method】In this experiment,broccoli tissue culture seedlings were acclimated for 7 days using liquid medium containing different concentrations of 75,100,150,200 mmol/L NaCl.The changes in physiological indicators and glucosinolate content of broccoli tissue culture seedlings under high salt stress(225 mmol/L NaCl)for 7 days were analyzed.And their physiological indicators were analyzed by membership function analysis to explore the physiological mechanism of the response of broccoli tissue culture seedlings to salt acclimation.【Result】The physiological indexes of broccoli tissue cultured seedlings had different degrees of response to salt acclimation.Salt acclimation significantly increased the POD activity,proline,soluble sugar,glucosinolates content of broccoli tissue culture seedlings compared to CK2(salt control),while the MDA content significantly decreased compared to CK1(blank control)and CK2.There was no significant difference in soluble protein content compared to CK2,while the total phenol and flavonoid content significantly decreased.The SOD activity in the 150 mmol/L NaCl acclimation group was significantly higher than that of CK2,at the same level as CK1.The anthocyanin content in the 200 mmol/L NaCl acclimation group was significantly higher than that of CK1 and CK2.The membership function analysis showed that the 150 mmol/L NaCl acclimation group had the highest comprehensive score of various physiological indicators under high salt stress.【Conclusion】Salt acclimation can improve the salt tolerance of broccoli tissue culture seedlings,and 150 mmol/L NaCl acclimation has the best effect.
