摘要
在筛选微生物源杀菌剂的过程中,从湖北省通山县九宫山采集的土样中分离到一株链霉菌,编号为HBERC-44441。经扫描电镜观察和16S rRNA基因序列比对,菌株被鉴定为利迪链霉菌Streptomyces lydicus。室内生物活性测定结果表明,该菌株发酵提取物对水稻纹枯病菌、小麦赤霉病菌、草莓灰霉病菌等具有强烈抑制活性。经分离制备,获得了其中几个活性组份,经波谱学分析,解析了其中4个化合物的化学结构,分别是吲哚-3-甲酸、pimprinine、另有两个pimprinine二聚体,鉴定为新的化合物。抗菌活性测试表明,吲哚-3-甲酸具有较好抗真菌活性,pimprinine及其二聚体抗真菌活性较弱,首次检测到pimprinine对植物重要病原细菌青枯劳尔氏菌有一定活性,其MIC值可达25μg/mL。
Streptomyces sp.HBERC-44441 was isolated from the soil sample collected from Jougongshan Mountain,Tongshan County,Hubei Province.Based on scanning electron microscopic(SEM)observation and 16S rRNA gene sequence analysis,the strain was identified as S.lydicus HBERC-44441.The bioassay results showed that the fermentation extracts of the strain exhibited excellent fungicidal activities against some important crop pathogenic fungi such as Rhizoctonia solani,Fusarium culmorum,Botrytis cinerea and so on.Several active compounds were purified by preparative HPLC,and the chemical structures of four compounds were elucidated by nuclear magnetic resonance(NMR)spectroscopy and high-resolution mass spectrometry(HRMS),they are indol-3-carboxylic acid,pimprinine,and two novel pimprinine dimers,respectively.The antimicrobial activity of pure compounds showed that pimprinine and its dimer had weak activity against some pathogenic fungi,while indol-3-carboxylic acid showed moderate activity.We found that pimprinine had moderate activity against important crop bacterial wilt for the first time,the MIC value is 25μg/mL.
