特医食品精优能缓解小鼠卵巢早衰发生的研究

摘要 目的探究以烟酰胺(NAM)和烟酸(NA)为主要有效成分的特医食品精优能对卵巢早衰(POF)的挽救作用。方法对3周龄ICR雌鼠进行单次白消安腹腔注射建立POF模型,对POF模型鼠连续灌胃28 d不同浓度的精优能,确定恰当的精优能处理用量(200 mg·kg^(-1)·d^(-1))。再按照处理不同将3周龄ICR雌鼠进行分组:空白对照组(CTRL组),给予PBS连续灌胃28 d;空白精优能处理组(C+200组),给予200 mg·kg^(-1)·d^(-1)精优能连续灌胃28 d;模型对照组(B组),白消安单次注射后,予PBS连续灌胃28 d;模型鼠精优能处理组(B+200组),白消安单次注射后,予200 mg·kg^(-1)·d^(-1)精优能连续灌胃28 d。各组处理28 d后,统计各组小鼠体重、卵巢指数、子宫指数;利用免疫组织化学染色法统计卵泡总数及各级卵泡比例;染色法采用卵母细胞体外培养技术统计卵子成熟率;并通过实时定量荧光PCR、蛋白质免疫印迹法检测卵巢内卵泡发育相关基因、蛋白及部分抗氧化酶的表达量。结果持续200 mg·kg^(-1)·d^(-1)的精优能灌胃28 d能够显著改善POF小鼠卵巢部分抗氧化酶(Sod2、Cat、Gpx4)的表达水平(P<0.05),显著改善POF小鼠的卵巢和子宫指数(P<0.05),显著增加卵泡数目(P<0.05),显著改善卵泡发育及卵巢储备相关基因(Gdf9、Bmp15、Amh)的表达水平(P<0.05),以及显著提高GV期卵母细胞数量和第一极体排出率(P<0.05)。结论精优能能够提高POF小鼠卵巢的抗氧化能力,维护卵巢储备与卵母细胞成熟,缓解POF的发展。 Objective:To explore the rescue effect of VigorKing,a special medical food with nicotinamide(NAM)and nicotinic acid(NA)as the main effective components,on the mice with premature ovarian failure(POF).Methods:POF model mice were established by a single intraperitoneal injection busulfan in three-week-old ICR mice.The mice were given continuous intragastric of different concentrations of VigorKing for 28 days to determine the appropriate concentration(200 mg·kg^(-1)·d^(-1))of VigorKing.Subsequently,the three-week-old ICR female mice were divided into four groups according to different treatments:administration of PBS continuously by gavage for 28 days in the blank control group(CTRL group);administration of 200 mg·kg^(-1)·d^(-1)VigorKing continuously by gavage for 28 days in the blank VigorKing treatment group(C+200 group);administration of PBS continuously by gavage for 28 days after a single injection of busulfan in model control group(group B);administration of 200 mg·kg^(-1)·d^(-1)VigorKing continuously by gavage for 28 days after a single injection of busulfan in the model rats in the model VigorKing treatment group(B+200 group).After the treatment for each group,the body weight,ovarian coefficient and uterine coefficient of mice in each group were analyzed.The total number of follicles and the proportion of follicles at different grades were determined by immunohistochemistry.The oocyte maturity rate was tested by oocyte culture in vitro.The expression levels of proteins related to follicle development and antioxidant enzyme genes in ovarian were detected by Western blot and real-time quantitative fluorescent PCR respectively.Results:Continuous gavage of 200 mg·kg^(-1)·d^(-1)VigorKing for 28 days significantly increased the expression levels of some antioxidant enzymes(Sod2,Cat,Gpx4)in the ovary of POF mice(P<0.05),significantly improved the ovarian and uterine indices(P<0.05),significantly increased the number of follicles effectively(P<0.05),and significantly enhanced the expression levels of genes related to follicle development and ovarian reserve(Gdf9,Bmp15,Amh)(P<0.05).The GV stage oocyte numbers and the first polar body extrusion(PBE)rate were also improved(P<0.05).Conclusions:VigorKing can improve ovarian antioxidant capacity of mice with POF,maintain ovarian reserve and oocyte maturation,and relieve the development of POF.
