白头翁汤对C57BL/6小鼠粪便中短链脂肪酸含量的影响 被引量:5

摘要 为了研究白头翁汤对正常小鼠粪便中肠道菌群代谢产物短链脂肪酸(SCFAs)含量的影响,本试验将24只清洁级C57BL/6小鼠随机分为空白组、白头翁汤低剂量组和白头翁汤高剂量组,每组8只,空白组灌胃生理盐水(0.9%),白头翁汤低、高剂量组分别灌胃白头翁汤2.7和8.1 g/(kg·bw·d)(按照生药量),同时每天测定小鼠体重;连续灌胃7 d后采集粪便,采用气相色谱法(GC)测定各组小鼠粪便中SCFAs(乙酸、丙酸、正丁酸、异丁酸、正戊酸和异戊酸)的含量并测量结肠长度。结果显示,与空白组相比,白头翁汤低剂量组和高剂量组的小鼠体重和结肠长度变化差异不显著(P>0.05);粪便中乙酸和丙酸含量均极显著升高(P<0.01),高剂量组正丁酸含量也显著升高(P<0.05);异丁酸、异戊酸、正戊酸含量变化不明显(P>0.05)。结果表明,白头翁汤对正常小鼠体重和结肠长度无影响,能够提高正常小鼠粪便中乙酸、丙酸和正丁酸含量。分析白头翁汤对正常小鼠粪便中SCFAs含量的影响可为白头翁汤的临床应用提供科学依据。 In order to investigate the effect of Pulsatilla decoction on the content of short-chain fatty acids(SCFAs)in the feces of normal mice,this study randomly devided 24 clean grade C57BL/6 mice into control group,low-dose Pulsatilla decoction group,and high-dose Pulsatilla decoction group,with 8 mice in each group.The control group was orally administered physiological saline(0.9%),while the low-dose and high-dose Pulsatilla decoction groups were orally administered 2.7 and 8.1 g/(kg·bw·d)of Pulsatilla decoction(according to the dosage of the herbal medicine),respectively.Body weights of the mice were measured daily,and after 7 consecutive days of administration,fecal samples were collected for gas chromatography(GC)analysis of SCFAs(acetic acid,propionic acid,N-butyric acid,isobutyric acid,N-valeric acid and isovaleric acid)contents,the lengths of the colons were also measured.The results showed that compared with the control group,there were no significant differences in body weight and colon length in the low-dose and high-dose Pulsatilla decoction groups(P>0.05),while the contents of acetic acid and propionic acid in the feces were extremely significantly increased(P<0.01)in both the low-dose and high-dose groups,and the content of N-butyric acid was significantly increased(P<0.05)in the high-dose group.There were no significant changes in the levels of isobutyric acid,isovaleric acid,and N-valeric acid(P>0.05).These findings suggest that Pulsatilla decoction has no effect on body weight and colon length in normal mice,but it can increase the contents of acetic acid,propionic acid,and N-butyric acid in the faces.Analyzing the effects of Pulsatilla decoction on SCFAs contents in the feces of normal mice provides scientific evidence for its clinical application.
