摘要
试验以甲基丙烯酸羟乙酯-甲基丙烯酸二甲氨乙酯[P(H-D)]承载盐酸左氧氟沙星(LVF),制备pH响应药物载体,然后制备改性聚甲基丙烯酸甲酯(PMMA)基复合骨水泥,并研究其性能和临床应用效果。结果表明:改性PMMA基复合骨水泥具备较好的pH响应释药性能,可以延缓释药周期,并且在吸水膨胀前后的抗压强度良好,分别为85.7、64.5 MPa,其面团时间和凝固时间分别为385.2、823.0 s,凝固温度为56.5℃,理化性能良好;在改性PMMA基复合骨水泥中添加3%交联剂N,N-亚甲基双丙烯酰胺(MBA)比较适宜;改性PMMA基复合骨水泥具备较好的细胞相容性和临床应用效果,均优于普通PMMA骨水泥。改性PMMA基复合骨水泥综合性能较好,可以在临床应用。
In this experiment,poly(Hydroxyethyl acrylate dimethylaminoethyl methacrylate)[P(H-D)]was used to carry Levofloxacin hydrochloride(LVF)to prepare pH responsive drug carrier,and then modified poly(Methyl methacrylate)(PMMA)was prepared based composite bone cement,and its performance and clinical application effectwere studied.The experimental results showed that the modified PMMA based composite bone cement had good pH responsive drug release performance,could delay the drug release cycle,and had good compressive strength before and after water absorption and expansion,which were 85.7 MPa and 64.5 MPa,respectively.Its dough time and setting time were 385.2 s and 823.0 s,respectively,and the setting temperature was 56.5℃.The physical and chemical properties were good.Adding 3%crosslinking agent N,N-methylenebisacrylamide(MBA)to modified PMMA based composite bone cement was more suitable.Modified PMMA based composite bone cement had good cell compatibility and clinical application effects,both of which were superior to ordinary PMMA bone cement.In summary,the modified PMMA based composite bone cement has good comprehensive performance and can be widely used in clinical practice.
