摘要
环磷酸鸟苷-腺苷酸合成酶(cyclic guanosine monophosphate synthase,cGAS)是重要的细胞质DNA模式识别受体,激活cGAS-STING信号通路进而诱导Ⅰ型干扰素(type I interferons,I-IFN)的表达,影响机体的免疫反应,在调节肿瘤免疫、病毒感染和自身免疫疾病方面发挥重要作用。通过cGAS-STING信号通路活化肿瘤微环境内的免疫细胞,启动抗肿瘤免疫应答已成为近年来的研究热点。cGAS-STING通路激动剂单独或联合其他治疗已应用于多种肿瘤的临床前研究。文章概述了cGAS-STING通路在肿瘤免疫调控中的研究进展及该通路在肿瘤免疫治疗中的临床应用。
Cyclic guanosine monophosphate synthase(cGAS)is an important cytoplasmic DNA pattern recognition receptor,which activates the cGAS-STING signaling pathway to induce the expression of type I interferons(I-IFN),affects the body's immune response and plays an important role in the regulation of tumor immunity,viral infections and autoimmune diseases.The activation of immune cells in the tumor microenvironment through cGAS-STING signaling pathway to initiate anti-tumor immune responses has become a hot topic of research in recent years.The agonists of cGAS-STING pathway alone or in combination with other therapies have been used in preclinical studies in a variety of tumors.This review outlines the research progress of cGAS-STING pathway in tumor immune regulation and the clinical applications in tumor immunotherapy.
