镉胁迫对泥鳅氧化毒性及肝组织损伤效应 被引量:1

Effects of cadmium stress on oxidative toxicity and liver histological damage in loach(Misgurnus anguillicaudatus)

摘要 将泥鳅置于4种不同浓度镉中饲养,分别在暴露10、20、30 d后,取雌、雄性泥鳅肝脏组织,采用邻苯三酚自氧化法、钼酸铵比色法检测超化物氧歧化酶(SOD)、过氧化氢酶(CAT)活性,采用H·E染色法观察肝脏组织结构变化。结果显示,雌性泥鳅在一定浓度Cd中暴露一定时间时,SOD活性显著升高,而雄性泥鳅在最低Cd浓度组暴露早期,SOD活性就极显著升高,并随Cd浓度增加呈先上升后下降的趋势;雌性泥鳅在最低Cd浓度组暴露早期,肝脏CAT活性就极显著升高,并随Cd浓度增大呈先升高后下降趋势,雄性泥鳅CAT活性变化趋势类似雌性,但变化程度不及雌鱼剧烈;泥鳅肝组织出现细胞间隙增大、细胞肿大、空泡化、细胞溶解、核溶解等现象,且雄性泥鳅损伤更为严重。说明一定剂量Cd对泥鳅可产生明显的氧化毒性效应,对雄性泥鳅SOD诱导作用较雌性强烈,而对雌性泥鳅CAT诱导作用又较雄性强烈;Cd胁迫可对泥鳅肝脏组织结构产生明显的损伤效应,其中雄性损伤较雌性更为严重。 Misgurnus anguillicaudatus were reared in four different concentrations of cadmium(0.005,0.05,0.5,5 mg/L).The liver tissues of female and male Misgurnus anguillicaudatus were taken after exposure for 10,20,30 days.The activities of superoxide dismutase(SOD)and catalase(CAT)were detected by pyrogallol autoxidation method and ammonium molybdate colorimetric method.The changes of liver tissue structure were observed by H·E staining.The results showed that the SOD activity of female loach increased significantly when exposed to a certain concentration of Cd for a certain time,while the SOD activity of male loach increased significantly at the early stage of exposure to the lowest Cd concentration group,and increased first and then decreased with the increase of Cd concentration.The CAT activity in the liver of female loach increased significantly at the early stage of exposure to the lowest Cd concentration group,and increased first and then decreased with the increase of Cd concentration.The change trend of CAT activity in male loach was similar to that of the female,but the degree of change was less than that of the female.The liver tissue of Misgurnus anguillicaudatus showed cell gap enlargement,cell swelling,vacuolization,cell dissolution,nuclear dissolution and other phenomena,and the damage of male Misgurnus anguillicaudatus was more serious.The above results suggest that a certain dose of Cd can produce obvious oxidative toxicity effect on loach.The induction of SOD in male loach is stronger than that in the female,and the induction of CAT in female loach is stronger than that in the male.Cd stress can cause obvious damage to the liver tissue structure of Misgurnus anguillicaudatus,and the damage of the male is more serious than that of the female.
