摘要
研究旨在优化坂口试剂改良法测定L-精氨酸发酵生产过程中的最佳工艺参数条件。以L-精氨酸为试验材料,考察不同检测波长、显色时间、pH值等条件对L-精氨酸含量测定结果的影响,探究检测方法的可靠性。结果显示,坂口试剂改良法的最佳检测条件为17.1 g/L氢氧化钠溶液7 mL、混合显色剂2 mL,离心并适当稀释的发酵液1 mL,30℃水浴30 min后测OD525 nm值,测定样品的加标回收率在99.20%~100.60%。研究表明,该方法的灵敏度高、重现性好、稳定性高,可对L-精氨酸发酵生产过程中的L-精氨酸含量进行准确、高效的检测。
The study was to optimize the optimal process parameters for determination of L-arginine in fermentation production process using an improved Sakuchi reagent method.Using L-arginine as test material,the effect of different detection wavelength,color development time,pH value and other conditions on determination of L-arginine content were investigated,and the reliability of detection method was explored.The results showed that the optimal detection conditions for the improved Sakuchi reagent method were 17.1 g/L sodium hydroxide solution in 7 mL,mixed chromogenic agent in 2 mL,centrifuged and appropriately diluted fermentation broth in 1 mL,water bath at 30℃for 30 min before measuring OD525 nm value.The measured recovery rate of the sample was between 99.20%and 100.60%.The study indicates that the method has high sensitivity,good reproducibility,high stability,and can accurately and efficiently detect the content of L-arginine inproduction process of L-arginine fermentation.
