Expression of SDC-1 in breast cancer and its clinical significance

摘要 目的 探究多配体蛋白聚糖-1 (Syndecans-1,SDC-1)在乳腺癌中的表达及其临床意义。方法 回顾性分析2021年1月至2021年6月本院收治的60例乳腺癌患者临床资料,收集乳腺癌患者术中切除的癌组织及经病理学检查未被浸润的癌旁正常乳腺组织,选用人正常乳腺上皮细胞系HBL-100和人乳腺癌上皮细胞系MCF-7,采用逆转录-聚合酶链式反应(RT-PCR)检测并比较HBL-100细胞和MCF-7细胞中SDC-1 mRNA相对表达量,采用Western Blot法检测并比较HBL-100细胞和MCF-7细胞中SDC-1蛋白表达量;采用免疫组织化学染色法检测并比较乳腺癌组织和癌旁正常组织中SDC-1阳性表达率,分析不同临床病理特征的乳腺癌患者癌组织中SDC-1阳性表达率,采用Spearman相关性分析SDC-1表达与乳腺癌临床病理特征的相关性。结果 人正常乳腺上皮细胞系HBL-100中SDC-1 mRNA、SDC-1蛋白相对表达量均高于人乳腺癌上皮细胞系MCF-7(P均<0.05);乳腺癌组织和癌旁正常组织中SDC-1表达情况差异显著,且乳腺癌组织中SDC-1阳性表达率低于癌旁正常组织(P均<0.05);不同年龄、病理类型乳腺癌患者的癌组织中SDC-1阳性表达率差异均不显著(P均>0.05),不同淋巴结转移情况、临床分期、肿瘤大小的乳腺癌患者癌组织中SDC-1阳性表达率水平均存在显著差异(P均<0.05);经Spearman相关分析,乳腺癌患者SDC-1表达与淋巴结转移情况、临床分期、肿瘤大小呈负相关(P均<0.05)。结论 SDC-1在乳腺癌组织中表达明显减弱,且其表达与乳腺癌淋巴结转移情况、临床分期、肿瘤大小负相关。 Objective To explore the expression of Syndecans-1(SDC-1) in breast cancer and its clinical significance.Methods The clinical data of 60 patients with breast cancer admitted to our hospital from January 2021 to June 2021 were retrospectively analyzed,and the cancer tissues removed during surgery of patients with breast cancer and the normal adjacent breast tissues that were not infiltrated by pathological examina-tion were collected.Human normal breast epithelial cell line HBL-100 and human breast cancer epithelial cell line MCF-7 were selected,and re-verse transcription-polymerase chain reaction(RT-PCR) were used to detect and compare the m RNA relative expression level of SDC-1 in HBL-100cells and MCF-7 cells and Western Blot for the protein expression level of SDC-1.Immunohistochemical staining was applied to detect the SDC-1positive expression rate in breast cancer tissues and normal adjacent tissues,and the positive expression rate of SDC-1 in cancer tissues of patients with breast cancer and different clinicopathological characteristics was analyzed.Spearman correlation analysis was used to analyze the correlation be-tween SDC-1 expression and clinicopathological characteristics of breast cancer.Results The relative expression levels of SDC-1 mRNA and SDC-1 protein in the human normal breast epithelial cell line HBL-100 were higher than those in the human breast cancer epithelial cell line MCF-7(all P<0.05).There was a significant difference in the expression of SDC-1 in breast cancer tissues and normal adjacent tissues,and the positive expression rate of SDC-1 in breast cancer tissues was lower than that in normal adjacent tissues(all P<0.05).There was no statistical difference in the SDC-1 positive expression rate in cancer tissues of patients with breast cancer of different ages and pathological types(all P>0.05),and there was a significant difference in the positive expression rate of SDC-1 in patients with breast cancer of different lymph node metastasis status,clinical stages and tumor sizes(all P<0.05).Spearman correlation analysis showed that the expression of SDC-1 in patients with breast cancer was negatively corre-lated with lymph node metastasis,clinical stage and tumor size(all P< 0.05).Conclusion The expression of SDC-1 is significantly reduced in breast cancer tissues,and its expression is negatively correlated with breast cancer lymph node metastasis,clinical stage and tumor size.
