胰腺癌不同分期的差异基因筛选及关键蛋白调控基因的预后分析 被引量:2

摘要 目的分析胰腺癌低分期组和高分期组差异表达基因和相关富集通路,结合预后信息探讨关键调控蛋白在胰腺癌发展中的作用,为更深入理解高分期组胰腺癌的分子机制提供理论支持。方法本研究使用TCGA数据库中不同肿瘤分期胰腺癌转录组数据来筛选差异表达基因,并对其进行功能富集分析和蛋白质互作网络构建。随后使用Cytoscape软件进行可视化和模块挖掘,最终对得到的关键蛋白调控基因进行生存分析。结果共筛选出差异基因277个,其中上调基因30个,下调基因247个。差异基因主要与消化作用、体液免疫反应、肠道胆固醇吸收及某些物质代谢过程等有关,与血液微粒和内质网腔等细胞组分有关,与丝氨酸相关酶活性、糖的结合和甘油三酸酯脂肪酶活性等分子功能有关。KEGG通路主要与胰腺分泌物、蛋白质消化吸收和脂肪消化吸收等有关。预后分析表明CPA1和AMY2B基因的表达水平与胰腺癌患者总生存时间显著相关(P<0.05)。结论关键蛋白调控基因CPA1和AMY2B在不同分期胰腺癌的发生和预后中紧密相关,为未来胰腺癌高分期患者精准治疗提供了新思路。 Objective To analyze the differentially expressed genes and related enriched pathways in the low-stage and high-stage groups of pancreatic cancer,and to explore the role of key regulatory proteins in the development of pancreatic cancer combined with prognostic information,so as to provide theoretical support for a deeper understanding of the molecular mechanism of pancreatic cancer in the high-stage group.Methods This study used the transcriptome data of pancreatic cancer with different tumor stages in the TCGA database to screen the differentially expressed genes,and performed functional enrichment analysis and protein-protein interaction network construction.Cytoscape software was then used for visualization and module mining to obtain key protein regulatory genes.Combined with the corresponding clinical information,the survival curves of key genes were drawn.Results A total of 277 differential genes were screened,including 30 up-regulated genes and 247 down-regulated genes.Differential genes were mainly related to digestion,humoral immune response,intestinal cholesterol absorption and metabolism processes of some substances,related to blood microparticles,endoplasmic reticulum lumen and other cell components,and related to molecular functions such as serinerelated enzyme activity,sugar binding and triglyceride lipase activity.The KEGG pathway was mainly related to pancreatic secretions,protein digestion and absorption,and fat digestion and absorption.Prognostic analysis showed that the expression levels of CPA1 and AMY2B genes were significantly correlated with the overall survival time of patients with pancreatic cancer(P<0.05).Conclusion The key protein regulatory genes CPA1 and AMY2B are closely related to the occurrence and prognosis of pancreatic cancer in different stages,providing new ideas for precise treatment of patients with high-stage pancreatic cancer in the future.
