黄芩多糖通过调节JAK 2/STAT 3通路和IL-23/IL-17炎性轴改善DSS诱导的UC模型小鼠的炎症 被引量:27

摘要 【目的】研究黄芩多糖对溃疡性结肠炎小鼠(UC)肠道炎症的影响及其机制。【方法】选用C57BL/6小鼠随机分为正常组、模型组、美沙拉嗪组以及黄芩多糖低、中、高剂量组(10只/组)。观察小鼠日常状态并记录疾病活动指数(DAI)评分。酶联免疫吸附测定(ELISA)法检测血清中细胞因子白细胞介素-6、17、23(IL-6、IL-17、IL-23)的表达。处死小鼠,HE观察小鼠肠黏膜损伤情况记录结肠组织损伤指数(TDI)评分,Western blot法和免疫组化法检测Janus激酶2(JAK2)、信号转导因子和转录激活因子(STAT3)、p-JAK2、p-STAT3蛋白的表达水平。【结果】与空白组相比,模型组小鼠DAI评分升高,血清中IL-6、IL-17、IL-23含量升高,结肠组织p-JAK2/JAK2、p-STAT3/STAT3比值升高,p-JAK2、p-STAT3表达量升高。与模型组相比,各给药组小鼠DAI评分降低;血清中IL-6、IL-17、IL-23含量降低,TDI评分降低,结肠组织p-JAK2/JAK2、p-STAT3/STAT3比值降低,p-JAK2、p-STAT3表达量降低。【结论】黄芩多糖可能通过JAK2/STAT3通路和IL-23/IL-17炎性轴改善UC小鼠的炎症。 【Objective】To investigate the effect and mechanism of Scutellaria Baicalensis polysaccharides on intestinal immunity in mice with ulcerative colitis(UC).【Methods】C57BL/6 mice were randomly assigned to blank group,model group,mesalazine group and Scutellaria Baicalensis polysaccharides low-dose,medium-dose and high-dose groups(n=10 mice/group).The daily status of mice was observed and the disease activity index(DAI)score was recorded.The expression of cytokines including IL-6,IL-17 and IL-23 in serum was detected by ELISA.After the mice were sacrificed,HE was used to observe the intestinal mucosal damage in mice,and the colonic tissue damage index(TDI)score was recorded.Western blot and immunohistochemistry were used to detect the expression levels of Janus kinase 2(JAK2),signal transducer and activator of transcription(STAT3),p-JAK2 and p-STAT3 proteins.【Results】Compared with the blank group,the DAI score of the model group was significantly increased,the contents of IL-6,IL-17 and IL-23 in serum were significantly increased,the ratio of p-JAK2/JAK2 and p-STAT3/STAT3 in colon tissue was significantly increased,and the expression levels of p-JAK 2 and p-STAT 3 were significantly increased.Compared with the model group,the DAI score of mice in each administration group was significantly decreased.The contents of IL-6,IL-17 and IL-23 in serum were decreased,the TDI score was decreased,the ratio of p-JAK 2/JAK 2 and p-STAT 3/STAT 3 in colon tissue was decreased,and the expression of p-JAK 2 and p-STAT 3 was decreased.【Conclusion】Scutellaria Baicalensis polysaccha-rides may improve the inflammatory effect of UC mice through JAK 2/STAT 3 pathway and IL-23/IL-17 inflammatory axis.
