岩陀黄酮与黄芩多糖复合物安全性评价 被引量:2

Safety Evaluation of The Compound of Rodgersia sambucifolia Hemsl. Flavonoids and Scutellaria baicalensis Georgi Polysaccharide

摘要 为探讨岩陀黄酮与黄芩多糖复合物的药用安全性,为其开发利用提供科学依据。开展了大鼠经口急性毒性试验、鼠伤寒沙门氏菌回复突变试验、小鼠精子畸形试验和骨髓细胞微核试验,对其进行安全性评价。结果显示,岩陀黄酮与黄芩多糖复合物对大鼠经口染毒半致死量(LD50)>5 g/kg·BW,为无毒级别,大鼠无临床症状和明显可见的病理变化;两次鼠伤寒沙门氏菌回复突变试验表明无致突变性;小鼠精子畸形率为1.99%~2.06%,高中低3个剂量组精子畸形率与阴性对照组比较差异不显著(P>0.05);骨髓细胞微核率高、中、低3个剂量组与阴性对照组比较差异不显著(P>0.05)。在本试验条件下,岩陀黄酮与黄芩多糖复合物属实际无毒物质,在2 500 mg/kg·BW内对试验动物未见急性毒性和遗传毒性。 The purpose of this paper was to study the medicinal safety of the compound of flavonoid from Rodgersia sambucifolia Hemsl. and polysaccharide from Scutellaria baicalensis Georgi, which will provide a scientific basis for its exploitation and utilization. Ames test, acute oral toxicity test(rats), Sperm aberration test(mice) and mice bone marrow micronucleus test were carried out to evaluate the safety of the complex of the two components mentioned above. The median lethal dose of the compound in acute oral toxicity test(rats) was LD50>5 g/kg·BW, which was non-toxic. The rats had no clinical symptoms or obvious pathological changes. There was no mutagenicity in two Salmonella typhimurium return mutation tests. The sperm distortion rate of mice was 1.99%~2.06%, and there was no significant difference between the three dosage groups and the negative control group(P>0.05). The micronucleus rate of bone marrow cells in high, medium and low dose groups was not significantly different from that in negative control group(P>0.05). In conclusion, under the condition of this experiment, the compounol of flavonoid from Rodgersia sambucifolia Hemsl. and polysaccharide from Scutellaria baicalensis Georgi was nontoxical substance, and there were no acute and genetic toxicity ant 2 500 mg/kg·BW.
