摘要
目的探讨核因子-κB(NF-κB)/基质金属蛋白酶9(MMP9)信号轴在姜黄素抑制卵巢癌SKOV3细胞增殖中的作用,为姜黄素的利用提供依据。方法体外培养卵巢癌SKOV3细胞,随机分为5μmol/L、10μmol/L、20μmol/L、40μmol/L姜黄素组和对照组,乳酸脱氢酶(LDH)实验检测姜黄素对SKOV3细胞毒性,MTT法检测卵巢癌SKOV3细胞的增殖情况。根据LDH和MTT实验结果,对10μmol/L、20μmol/L姜黄素组和对照组进行进一步分析,RT-PCR和Western blot检测卵巢癌SKOV3细胞中NF-κB、MMP9 mRNA和蛋白相对表达水平。结果随着剂量的增加,姜黄素对SKOV3细胞的毒性增强,并具有剂量依赖性,差异有统计学意义(P<0.05)。与对照组相比,随着剂量的增加,姜黄素可以显著降低卵巢癌SKOV3细胞的存活率,并具有剂量依赖性,差异有统计学意义(P<0.05)。姜黄素可以显著降低SKOV3细胞中NF-κB和MMP9 mRNA相对表达水平,降低SKOV3细胞中p-NF-κB和MMP9蛋白表达水平,差异有统计学意义(P<0.05)。结论姜黄素能够有效抑制卵巢癌SKOV3细胞的增殖,NF-κB/MMP9信号轴在此过程中起到重要的调节作用。
Objective To investigate the role of NF-κB/MMP9 signal axis in curcumin inhibiting the proliferation of ovarian cancer SKOV3 cells,provide evidence for the utilization of curcumin.Methods Ovarian cancer SKOV3 cells were cultured in vitro,randomly divided into 5μmol/L,10μmol/L,20μmol/L,40μmol/L curcumin group and control group,the lactate dehydrogenase(LDH)assay was used to detect the toxicity of SKOV3 cells,MTT assay was used to detect the proliferation of ovarian cancer SKOV3 cells.According to the results of LDH experiment and MTT experiment,further analysis was conducted on the 10μmol/L,20μmol/L curcumin group and control group,and the relative expression levels of NF-κB and MMP9 mRNA and protein in SKOV3 cells were detected using by RT-PCR and Western blot.Results With the increase of dose,the toxicity of curcumin on SKOV3 cells increased in a dose-dependent manner,the difference was statistically significant(P<0.05).Compared with the control group,with the increase of dose,curcumin could significantly reduce the survival rate of ovarian cancer SKOV3 cells in a dose-dependent manner,the difference was statistically significant(P<0.05).Curcumin could significantly reduce the relative expression level of NF-κB and MMP9 protein in SKOV3 cells,and reduce the expression level of p-NF-κB and MMP9 protein in SKOV3 cells.the difference was statistically significant(P<0.05).Conclusion Curcumin can effectively inhibit the proliferation of ovarian cancer SKOV3 cells,the NF-κB/MMP9 signal axis plays an important role in this process.
