摘要
目的探讨输合配穴针刺法对小儿缺氧缺血性脑病(hypoxic-ischemic brain damage,HIBD)模型大鼠Nrf2/ARE信号通路的影响。方法将120只健康新生大鼠随机分为输合配穴针刺组、模型组和空白对照组,再将各个组的新生大鼠平均分成4个时间小组即为0、3、7、14 d,每个小组10只新生大鼠。实验采用颈动脉结扎法建立缺血缺氧性脑病模型。输合配穴针刺组于造模成功后给予新生大鼠患侧输合配穴针刺法进行针刺治疗,模型组对新生大鼠仅进行造模操作,空白对照组不对新生大鼠做任何处理。实验采用Western blot方法和RT-PCR检测法对新生大鼠脑组织中Nrf2、Keap1和HO-1的蛋白表达情况及mRNA相对表达量进行检测并对结果进行对比分析。结果模型组于造模7 d后Nrf2、Keap1和HO-1蛋白表达和mRNA相对表达量最多,差异具有统计学意义(P<0.01)。输合配穴针刺组中与相对应时间点的模型组对比,都明显高于模型组,差异具有统计学意义(P<0.01)。说明输合配穴针刺法对HIBD模型Nrf2/ARE信号传导通路的是有影响的。结论输合配穴针刺法通过促进Nrf2/ARE信号通路上的关键蛋白因子Nrf2的表达,在细胞中与Keap1解离后转移至细胞核,从而激活下游蛋白因子HO-1的表达,减轻HIBD对机体的氧化损伤,以达到保护脑细胞和脑组织的目的。
Objective To explore the effect of Shu-He acupoints combination acupuncture on Nrf2/ARE signal transduction pathway in hypoxic-ischemic brain damage(HIBD)model.Methods A total of 120 healthy newborn rats were randomly divided into Shu-He acupoints combination acupuncture group,model group and blank control group.The newborn rats in each group were divided into four time groups,namely 0 day,3 days,7 days and 14 days groups.There were 10 newborn rats in each group.The model of hypoxic ischemic encephalopathy was established by carotid artery ligation.After the success of modeling,the Shu-He acupoints combination acupuncture group was given acupuncture treatment on the affected side of neonatal rats.The model group only made modeling operation on neonatal rats,and the blank control group did not do any treatment on neonatal rats.In this experiment,the protein expression and relative mRNA expressions of Nrf2,Keap1 and HO-1 in neonatal rat brain were detected by Western blot and RT-PCR,and the results were compared and analyzed.Results The relative expressions of Nrf2,Keap1 and HO-1 protein and mRNA were the most in the model group 7 days after modeling(P<0.01).Compared with those of the model group at the corresponding time point,the expressions of the Shu-He acupoints combination acupuncture group was significantly higher,and the difference was statistically significant(P<0.01).It showed that the Shu-He acupoints combination acupuncture has an effect on the expression of Nrf2/ARE signal transduction pathway in HIBD model.Conclusion By promoting the expression of the key protein factor Nrf2 on the Nrf2/are signal pathway,Shu-He acupoints combination acupuncture method dissociates with Keap1 in cells and transfers to the nucleus,so as to activate the expression of downstream protein factor HO-1 and reduce the oxidative damage of HIBD to the body,so as to protect brain cells and brain tissues.
