壳聚糖浸种对高山杜鹃种子萌发及生长的影响研究 被引量:4

摘要 以秦岭高山杜鹃为研究对象,以不同浓度CTS(0.05%、0.10%、0.15%、0.20%、0.25%、0.30%)作为引发剂对高山杜鹃浸种处理后种子萌发指标、幼苗生化指标的影响进行了分析,为CTS应用到杜鹃种子的萌发、克服高山杜鹃种子萌发率低、种间差异大以及引种驯化等问题提供了解决途径,并为其扩大培育提供理论参考。结果表明:在0.15%CTS浸种条件下,高山杜鹃的发芽势、发芽率、发芽指数均达到峰值,处理效果最佳,与CK相比分别提高了25个百分点、30个百分点和46%;高山杜鹃幼苗的保护酶SOD、POD、CAT活性与CK相比分别提高了89%、164%、64%;叶绿素含量提高了64%;可溶性蛋白含量提高了92%,且经单因素方差分析显示,以上各生化指标受外源CTS处理的影响显著。综上所述,0.15%CTS溶液作为引发剂能够显著提高种子的代谢水平,加速细胞内物质的转化,提高种子的萌发率;高山杜鹃幼苗喷洒0.15%CTS溶液后,其抗氧化物酶活性、叶绿素含量均呈现明显的上升趋势,植物的抗逆性也得到提高,缓解了因膜脂过氧化程度过高对植物机体造成的氧化损伤,促进了光合产物的积累。 In order to overcome the problems of low seed germination rate of Rhododendron lapponicum,large inter-specific differences,and introduction and domestication,and provide theoretical reference for its expansion cultivation,the effects of different concentrations of chitosan CTS(0.05%,0.10%,0.15%,0.20%,0.25%and 0.30%)as initiator soaking on seed germination index and seedling biochemical index of Rhododendron lapponicum from Qinling were studied,The results showed that under the condition of 0.15%CTS soaking,the germination potential,germination rate and germination index of Rhododendron lapponicum reached the peak value,and the treatment effect was the best,which increased by 25 percentage points,30 percentage points and 46%,respectively,compared with CK.Compared with the control,the activities of protective enzymes SOD,POD and CAT in Rhododendron lapponicum seedlings increased by 89%,164%and 64%,respectively.Chlorophyll content increased by 64%,and Soluble protein content increased by 92%.One-way analysis of variance showed that the above physiological indexes were significantly affected by exogenous chitosan treatment.In conclusion,0.15%CTS solution as initiator could significantly improve the metabolism level of seeds,accelerate the transformation of intracellular substances,and improve the germination rate of seeds.After spraying 0.15%CTS solution on Rhododendron lapponicum seedlings,their antioxidant enzyme activity and chlorophyll content showed an obvious increasing trend.At this time,the stress resistance of plants was enhanced to alleviate the oxidative damage caused by excessive membrane lipid peroxidation degree on the plant body and increase the accumulation of photosynthates.
