摘要
目的探索硫酸锌是否通过抑制活性氧促进脂多糖(lipopolysaccharides,LPS)诱导的小胶质细胞向M2型极化。方法提取小鼠原代小胶质细胞,应用3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐(MTT)比色法确定各药品的最适浓度;通过免疫印迹方法检测各组细胞中IL-1β和Arg-1蛋白表达差异;应用qRT-PCR技术检测各组细胞中白介素-1β(interleukin-1β,IL-1β)mRNA和精氨酸酶1(arginase-1,Arg-1)mRNA的表达差异。结果LPS显著增加IL-1β蛋白和mRNA的表达,诱导小胶质细胞向M1型转化。硫酸锌减少IL-1β蛋白和mRNA的表达,增加Arg-1蛋白和mRNA的表达,促进LPS诱导的小胶质细胞向M2型转化,但是此作用被H_(2)O_(2)逆转。结论硫酸锌可通过抑制活性氧促进LPS诱导的M1型小胶质细胞向M2型极化。
Objective To explore whether the microglia treated with zinc sulfate promotes the transformation of M2 phenotype through influencing oxidative stress.Methods Passage microglia of the mice was extracted,and the most applicable concentration of each drug was detected with MTT colorimetric method.The differences in the level of IL-1βand Arg-1 in the cells of each group were detected by Western blotting.qRT-PCR was used to detected the differences of IL-1βmRNA and Arg-1 mRNA.Results The expression of IL-1βof microglia significantly increased by LPS,and LPS could inducing microglia transformation to M1 type.Zinc sulfate can decrease the expressions of IL-1βprotein and mRNA and increase the expressions of Arg-1 protein and mRNA.Zinc sulfate can promote the transformation of M1 microglia induced by LPS to M2,which can be reversed by H_(2)O_(2).Conclusion Zinc sulfate can promote polarization of LPS-induced microglia to M2 phenotype by inhibiting reactive oxygen species.
