摘要
细胞是生命体最基本的功能单元。传统均一性的群体研究理念难以满足对细胞个体特征的分析需求,只有在单细胞层面认识细胞的异质性,才能深入理解生命活动的本质。与单细胞基因组和转录组相比,单细胞蛋白质组学分析目前尚处于起步阶段。由于蛋白质无法像基因进行扩增,因此,单细胞蛋白质组的规模化分析仍面临巨大挑战。本文对现有的单细胞蛋白质组分析技术进行了归纳和总结,重点介绍了基于质谱的单细胞蛋白质组学在样品处理技术以及定量分析方法方面的最新进展,并对单细胞蛋白质组学分析技术的发展趋势进行了展望。
Cells are the most fundamental function units in living organisms.However,it is difficult to obtain unique characteristics of individual cells by the traditional omics analysis of bulk population cells.Therefore,to understand the essence of life activities,it is essential to perform single cell analysis to recognize cell heterogeneity.Compared with single-cell genomes and transcriptomes,currently single-cell proteomics is still in infancy,and the large-scale proteome analysis in single cells still faces great challenges since proteins cannot be amplified like genes.In this review,we summaried the existing mass spectrometry based single cell proteome analytical technologies,focusing on the latest progress in sample processing techniques and quantitative proteome analysis.Finally,the trends in development of single-cell proteomics analysis technologies were also prospected.
